Summary: Subarachnoid hemorrhage (SAH) is the third most common subtype of stroke. The incidence has decreased over the years, possibly due to lifestyle changes. A quarter of SAH patients die before hospital admission, and those admitted to the hospital have improved outcomes but may experience long-term neuropsychiatric complications. Severe sudden headache is the most common symptom, and diagnosis is typically made through CT scan and angiography. Treatment focuses on reducing the risk of rebleeding and early aneurysm treatment.

Summary: Transplantation of stem cell-derived retinal pigment epithelial (RPE) cells is a potential treatment for age-related macular degeneration (AMD). However, there is limited understanding of how the recipient retina regulates the survival and maturation of transplanted RPE cells. In this study, we transplanted stem cell-derived RPE into rabbits and analyzed their gene expression, finding that the transplanted RPE cells retained their identity and showed maturation towards the normal RPE state. Transcription factors FOS, JUND, and MAFF were activated in the transplanted RPE cells and played important roles in regulating gene expression related to supporting host photoreceptor function and promoting cell survival.

This Primer provides a comprehensive overview of the epidemiology, pathophysiology, diagnosis, and treatment of ICH, highlighting the importance of prevention and management of haematoma expansion and long-term vascular risk.

Summary: This study investigated the clinical/imaging features for identifying NOTCH3-mutation-related intracerebral haemorrhage (ICH) in a cohort of 749 cerebral small vessel disease (CSVD) patients. The study found that patients with NOTCH3 mutation were more likely to have thalamic haemorrhage, a family history of stroke and more severe CSVD neuroimaging markers. A NOTCH3-ICH score was developed to assist in identifying genetic causes of ICH.

Summary: This study conducted single-cell transcriptomic profiling of an hESC-RPE differentiation protocol for treating AMD and found that the cells exhibited characteristics of early embryonic development during differentiation, ultimately obtaining a pure RPE cell population. The findings of this study are significant for the development of safe and effective pluripotent stem cell-based therapies for AMD.

Summary: This study demonstrates the neurotoxic effects of Pamiparib on zebrafish, including cerebral hemorrhage, brain atrophy, and movement disorders. Pamiparib exposure leads to downregulation of acetylcholinesterase and adenosine triphosphate activities, upregulation of oxidative stress, apoptosis, and disruption of gene expression involved in neurodevelopment, neurotransmitter pathways, and Parkinson's disease-like symptoms. Astaxanthin and activation of Notch signaling can partially rescue the neurodevelopmental defects caused by Pamiparib.
