Reduction of renal uptake of 111In-DOTA-labeled and A700-labeled RAFT-RGD during integrin αvβ3 targeting using single photon emission computed tomography and optical imaging

Integrin av beta 3 expression is upregulated during tumor growth and invasion in newly formed endothelial cells in tumor neovasculature and in some tumor cells. A tetrameric RGD-based peptide, regioselectively addressable functionalized template-(cyclo-[RGDfK])4 (RAFT-RGD), specifically targets integrin av beta 3 in vitro and in vivo. When labeled with indium-111, the RAFT-RGD is partially reabsorbed and trapped in the kidneys, limiting its use for further internal targeted radiotherapy and imaging investigations. We studied the effect of Gelofusine on RAFT-RGD renal retention in tumor-bearing mice. Mice were imaged using single photon emission computed tomography and optical imaging 1 and 24 h following tracer injection. Distribution of RAFT-RGD was further investigated by tissue removal and direct counting of the tracer. Kidney sections were analyzed by confocal microscopy. Gelofusine significantly induced a >50% reduction of the renal reabsorption of 111In-DOTA-RAFT-RGD and A700-RAFT-RGD, without affecting tumor uptake. Injection of Gelofusine significantly reduced the renal retention of labeled RAFT-RGD, while increasing the tumor over healthy tissue ratio. Summary: Aza-boron-dipyrromethenes (Aza-BODIPYs) are a class of fluorophores with azadipyrromethene (aza-DIPY) ligand rigidified by a boron atom. Complexes of group 13 metals (aluminum and gallium) were synthesized and their properties were investigated. AzaGaDIPY and azaAlDIPY exhibited significant red-shifted fluorescence compared to aza-BODIPY, making them promising for optical medical imaging.

Summary: This study developed hydrophobic photoluminescent polymeric nanoparticles and investigated their interaction with macrophages. Multimeric RGD-Based Strategies for Selective Drug Delivery to Tumor Tissues

Summary: RGD peptides have gained significant attention in the past two decades for their potential in improving tumor therapy by targeting the alpha V beta 3 integrin receptor. This review focuses on the molecular design of multimeric RGD compounds and suitable linkers for drug delivery. The use of RGD constructs in enhancing binding affinity and drug uptake in tumor cells, as well as their potential as theranostic systems, is highlighted. Furthermore, ongoing and future development in comparison with large molecules like monoclonal antibodies is addressed.

Summary: Using fluorescence-guided surgery (FGS) with NIR-II dyes targeting human epidermal growth factor receptor 2 (HER2)-positive ovarian tumors showed favorable tumor accumulation and selective targeting of HER2 positive tumors. These bioconjugated NIR-II-emitting dyes displayed prolonged stability in serum and maintained affinity toward HER2 in vitro, indicating their potential for NIR-II FGS in the cancer setting. This study suggests that the use of NIR-II dyes in FGS can improve the complete resection of microscopic ovarian tumors.

Molecular imaging of liver inflammation using an anti-VCAM-1 nanobody

Summary: In this study, researchers found that molecular imaging of VCAM-1 could be a non-invasive alternative to biopsy for detecting liver inflammation in chronic liver disease. By using radiolabeled anti-VCAM-1 nanobody and SPECT, they were able to quantify liver uptake and correlate it with histological inflammatory scores and molecular markers. The diagnostic power of this imaging tool to detect liver inflammation was excellent, suggesting its potential for clinical use.

Summary: In this study, ultra-small gold nanoparticles stabilized by co-ligands were synthesized, and their physicochemical properties were controlled by the amount of reducing agent used. The absorption cross-section was significantly increased by precisely controlling the reducing agent, which in turn influenced the photoluminescence quantum yield. The results suggest that the physicochemical properties of the ligand shell can be tuned to control the near-infrared absorption and photoluminescence of the gold nanoparticles.
