Antitumor activities of valproic acid on Epstein-Barr virus-associated T and natural killer lymphoma cells

EpsteinBarr virus (EBV), which infects B cells, T cells, and natural killer (NK) cells, is associated with multiple lymphoid malignancies. Recently, histone deacetylase (HDAC) inhibitors have been reported to have anticancer effects against various tumor cells. In the present study, we evaluated the killing effect of valproic acid (VPA), which acts as an HDAC inhibitor, on EBV-positive and -negative T and NK lymphoma cells. Treatment of multiple T and NK cell lines (SNT13, SNT16, Jurkat, SNK6, KAI3 and KHYG1) with 0.1-5 mM of VPA inhibited HDAC, increased acetylated histone levels and reduced cell viability. No significant differences were seen between EBV-positive and -negative cell lines. Although VPA induced apoptosis in some T and NK cell lines (SNT16, Jurkat and KHYG1) and cell cycle arrest, it did not induce lytic infection in EBV-positive T or NK cell lines. Because the killing effect of VPA was modest (1 mM VPA reduced cell viability by between 22% and 56%), we tested the effects of the combination of 1 mM of VPA and 0.01 mu M of the proteasome inhibitor bortezomib. The combined treated of cells with VPA and bortezomib had an additive killing effect. Finally, we administered VPA to peripheral blood mononuclear cells from three patients with EBV-associated T or NK lymphoproliferative diseases. In these studies, VPA had a greater killing effect against EBV-infected cells than uninfected cells, and the effect was increased when VPA was combined with bortezomib. These results indicate that VPA has antitumor effects on T and NK lymphoma cells and that VPA and bortezomib may have synergistic effects, irrespective of the presence of EBV. Gait performance and dual-task costs in school-aged children with Down syndrome

Summary: This study aimed to assess gait performance and its correlation with physical functions in children with Down syndrome (DS), as well as the dual-task costs. The results showed that children with DS had poorer balance function, muscle strength, and gait quality compared to the control group. In the DS group, there was a positive correlation between gait speed, step length, and intelligence quotient. Dual-task costs for gait speed, step length, and cadence were greater in the DS group, highlighting the importance of appropriate interventions for motor functions in these children.

Decreased Walking Efficiency in Elementary School Children with Developmental Coordination Disorder Trait

Summary: The objective of this study was to compare walking efficiency and associated physical function between children with and without developmental coordination disorder trait. The study found that compared to the control group, the developmental coordination disorder group had lower walking efficiency, higher step length variability, and shorter single leg stance test. These results suggest that developmental coordination disorder trait can affect walking efficiency and physical function in children.

Pathological gait in Rett syndrome: Quantitative evaluation using three-dimensional gait analysis

Summary: In this study, we quantitatively evaluated gait pathology in patients with Rett syndrome using three-dimensional gait analysis. The results showed that patients with Rett syndrome exhibit abnormal gait, especially in the coronal plane of the pelvis and hip joint, as well as the horizontal plane of the pelvis.

N6-methyladenosine Modification of Hepatitis B Virus RNA in the Coding Region of HBx

Summary: N6-methyladenosine (m(6)A) is a post-transcriptional RNA modification involved in various processes including transport, degradation, translation, and splicing. The study finds that HBV RNA is predominantly modified by m(6)A in the coding region of HBx. Mutations in the methylation sites reduce HBV mRNA and HBs protein levels. Inhibiting or knocking down m(6)A in primary hepatocytes decreases viral RNA and HBs protein levels in the medium, indicating the importance of m(6)A modification for efficient HBV replication in hepatocytes.

Summary: A 32-year-old woman with chronic active Epstein-Barr virus infection experienced graft failure after her first allogeneic hematopoietic cell transplant. She then underwent a second transplant using haploidentical hematopoietic cells and posttransplant cyclophosphamide as salvage therapy. Neutrophils engrafted successfully on day 19, and the patient remained in remission without severe transplant-related complications for 10 months.

Diagnostic and disease severity determination criteria for hydroa vacciniforme lymphoproliferative disorders and severe mosquito bite allergy

Summary: Hydroa vacciniforme lymphoproliferative disorder (HV-LPD) and severe mosquito bite allergy (SMBA) are cutaneous forms of Epstein-Barr virus (EBV)-associated T/natural killer (NK) cell LPDs. They are closely related to chronic active EBV disease (CAEBV) and EBV-associated hemophagocytic lymphohistiocytosis (EBV-HLH). Diagnostic criteria and determination criteria for disease severity are proposed to define the disease spectrum of HV-LPD and SMBA. The proposed diagnostic criteria are in line with the guidelines for the management of CAEBV and related disorders 2023.

Droplet Digital PCR Development for Adenovirus Load Monitoring in Children after Hematopoietic Stem Cell Transplantation

Summary: In this study, a novel pathogen detection technology called droplet digital PCR (ddPCR) was developed and compared with real-time quantitative PCR (qPCR) for detecting human adenovirus DNA. The results showed that ddPCR exhibited better reproducibility and sensitivity, as well as equivalent quantifiability, compared with qPCR. Therefore, ddPCR has the potential to provide a more accurate DNAemia detection, determine cutoff values for treatment initiation, and enable antiviral efficacy assessment.

Epstein-Barr Virus BBLF1 Mediates Secretory Vesicle Transport to Facilitate Mature Virion Release

Summary: Studies have shown that secretory vesicles derived from the trans-Golgi network or endosomes transport virions into the extracellular space, but the mechanism underlying the release of Epstein-Barr virus, a human oncovirus, remains unclear. In this study, we found that disruption of BBLF1, a tegument component, suppressed viral release and resulted in the accumulation of viral particles on the inner side of the vesicular membrane. Our findings suggest that BBLF1 regulates the viral release pathway and reveal a new aspect of tegument protein function.

The results showed that nanopore sequencing could detect RNA viral pathogens with equivalent sensitivity and genome coverage as Illumina sequencing, and it had the advantage of shorter sequencing time.

Growth Transformation of B Cells by Epstein-Barr Virus Requires IMPDH2 Induction and Nucleolar Hypertrophy

Summary: In this study, EBV infection was found to induce the expression of the IMPDH2 gene, leading to nucleolar, nuclear, and cellular hypertrophy, as well as efficient cell proliferation. The use of an IMPDH2 inhibitor, MMF, was shown to suppress PTLD. These findings provide foundational evidence for understanding the mechanisms of B cell transformation induced by EBV.

Evaluation of Muscle Oxygen Dynamics in Children's Gait and Its Relationship with the Physiological Cost Index

Summary: This study aimed to evaluate muscle oxygen saturation in lower extremity muscles during walking in children, its changes with age, and the relationship between the physiological cost index. Results showed that changes in muscle oxygen saturation were greater in the lower leg muscles, particularly the tibialis anterior and medial head of the gastrocnemius. The physiological cost index was significantly correlated with changes in muscle oxygen saturation in the tibialis anterior, suggesting a potential influence on walking efficiency.
