Summary: This study aimed to investigate the prognosis of completely resected embryonal RMS at sites other than paratesticular, uterine, or vaginal primary sites. The results showed that patients with nonalveolar RMS completely resected at diagnosis have excellent survival rates when arising from nonparatesticular, uterine, and vaginal sites, and can be successfully treated with low-intensity chemotherapy.

Summary: The updated results of the RMS2005 trial confirmed that adding maintenance chemotherapy (MC) with vinorelbine and low dose cyclophosphamide to standard treatment improved survival in patients with non-metastatic high risk rhabdomyosarcoma. However, a more recent study using different drugs in the MC phase did not show a survival improvement, suggesting that not all types of MC are equally effective. More investigations are needed to determine the role of MC in metastatic or relapsed RMS.

Summary: This study found that combined treatment of D,L-methadone with doxorubicin, carboplatin, and vincristine had strong antitumor effects on pediatric rhabdomyosarcoma (RMS) cell lines. These effects were mediated by increased ROS production and up-regulation of caspase-3/7 activity. The combination of D,L-methadone with doxorubicin significantly reduced cell migration in both cell lines.

Summary: Esophageal rhabdomyosarcoma is a rare aggressive neoplasm with high malignancy and metastatic potential. Patients often present at a late stage, leading to poor prognosis. Combination therapy involving radiotherapy and chemotherapy can provide short-term symptom relief, but long-term survival rates remain low.

Summary: Clinical trial eligibility criteria play a crucial role in controlling patient characteristics, ensuring participant safety, and optimizing result interpretation. However, complex and overly restrictive criteria have posed barriers to patient access, recruitment, and the generalizability of trial results. In response, a working group developed a framework for lung cancer clinical trial eligibility criteria, aiming to simplify criteria, facilitate stakeholder search for trials, and harmonize trial populations for treatment effect comparisons.

Summary: The study examined meniscal repair surgeries in pediatric and adolescent patients across multiple institutions. The results showed that higher BMI was associated with complex and radial tear repairs, while lateral tears were more common in male and younger patients. Further studies should investigate factors related to meniscal repair survival and outcomes in this pediatric cohort undergoing knee surgery.

Summary: Children and adolescents exhibit high variability in brain development, with females generally passing through developmental stages earlier than males by up to 1 year. The difference between brain age and chronological age is heritable, as is the change in this difference over time. Reliable brain age predictors can help detect early signs of abnormal brain development and potential risk for neurodevelopmental disorders.

Summary: Plant-extracted essential oils are considered as potential alternatives to synthetic insecticides for insect pest control. Although they are generally perceived as safe for the environment and non-target organisms, their unintended effects on insect biological and reproductive traits have been overlooked. This study examined the toxicity of Eucalyptus globulus essential oil to Drosophila suzukii and found that low concentrations of the oil enhanced the fecundity and lifespan of the flies in a sex- and mating status-dependent manner. These findings highlight the need for further investigation and consideration of the age, sex, and mating status-dependent responses to low concentration exposures of essential oils.

Summary: The outcomes of patients with alveolar rhabdomyosarcoma N1 were similar in both protocols, but patients with FOXO1 fusion-negative tumors enrolled in RMS2005 showed significantly better outcomes, indicating that different chemotherapy strategies may impact the outcome of this subgroup of patients.

Summary: This study found that about 1.2% of boys reported trading sex, and these boys were more likely to identify as Black or Indigenous, have a marginalized gender identity or sexual expression, have engaged in sexual intercourse, and have experienced various forms of destabilizing circumstances including homelessness, food insecurity, foster care, and substance treatment. These findings challenge assumptions and stereotypes and open up new avenues for research, intervention, and prevention.

Summary: This study examined the long-term neurocognitive outcomes of survivors of childhood osteosarcoma and Ewing sarcoma. The results showed that these survivors are more likely to report neurocognitive difficulties, which are related to employment status and chronic health conditions. Therefore, early screening, prevention, and treatment of chronic health conditions may be important for improving/preventing long-term neurocognitive outcomes.

Summary: Disparities in care, treatment-related toxicity, and health-related quality of life for adolescents and young adults with cancer are often disregarded due to limited collection of patient-reported outcomes in cancer clinical trials. Standardizing the assessment of HRQoL and treatment toxicity in AYA CCTs is crucial for improving patient outcomes. The National Cancer Institute's Clinical Trials Network AYA PRO Task Force has identified key considerations and selected appropriate measures to address this issue.

Summary: This study explored patient-reported outcomes in head and neck rhabdomyosarcoma (HNRMS) survivors and assessed the relationship between physician grading of adverse events (AE) and patient reporting. The findings highlight the importance of incorporating patient-reported outcome measures in survivorship follow-up, as physician-graded AEs are not sufficient to provide tailored care for HNRMS survivors.

Summary: This study analyzed the medical records of 59 patients with 64 lesions of benign intracranial meningioma who were treated with proton therapy. With a median follow-up of 6.3 years, the 5-year rates of local progression and grade 3 or greater toxicity were 6% and 2% respectively. The 5-year overall survival rate was 87%. In conclusion, fractionated proton therapy up to 50.4 GyRBE is a safe and highly effective treatment for benign intracranial meningioma.

Summary: Proton beam therapy (PBT) is an advanced radiotherapy technique that has seen exponential growth in demand and capacity worldwide. However, there are geographic inequalities in the distribution of PBT centers, resulting in variations in access and use. This study aims to identify the factors contributing to these inequalities and raise awareness among stakeholders and policy makers. The findings highlight socioeconomic status and geographical location as key indicators of disparity in access to PBT, and further research is needed to reduce the care gap and address ethical considerations, particularly for pediatric patients.

Summary: This study aims to quantify facial deformation in survivors of head and neck rhabdomyosarcoma (HNRMS) and investigate the differences between different treatment modalities. The findings show that HNRMS survivors have significantly reduced facial growth and increased facial asymmetry compared to healthy controls, regardless of treatment modality. Furthermore, there is significantly more facial deformation in orbital patients when comparing radiation therapy (RT) to surgery with brachytherapy (AMORE).

Summary: This systematic review examines the use of proton beam therapy (PBT) in the management of recurrent central nervous system (CNS) and base of skull (BoS) tumors. The findings suggest that PBT can provide good local control with acceptable toxicity rates in the reirradiation setting. However, there are several factors to consider that may increase the risk of toxicities in CNS or BoS tumor recurrences, highlighting the importance of patient selection.

Summary: This study aimed to determine the prevalence of pulmonary nodules cavitation in pediatric and young adult patients with sarcoma undergoing tyrosine kinase inhibitor therapy, and to assess whether cavitation can predict clinical response and survival outcomes. The results showed that during tyrosine kinase inhibitor therapy, some patients had cavitation in their lung nodules. The occurrence of cavitation was unrelated to disease progression and significantly correlated with longer progression-free survival in sarcoma patients.
