Background: This study was designed to evaluate the prognostic value of tumour stage T2 subcategorization (T2a and T2b) in patients with gastric carcinoma. Methods: Clinicopathological details of a prospective series of patients who had radical resection of gastric adenocarcinoma in a single institution were analysed. Univariable and multivariable survival analyses were performed with the log rank test and Cox's model respectively. Results: Of 373 evaluable patients, 49 (13.1 per cent) had a T2a and 143 (38.3 per cent) a T2b tumour. At a median follow-up of 35.5 months, the 5-year overall survival rate was 73 and 31.1 percent for patients with T2a and T2b lesions respectively (P < 0.001). On multivariable analysis, T stage remained an independent prognostic factor. Compared with T1a, the mortality risk for patients with T1b (hazard ratio (HR) 1.00; P = 0.992) and T2a (HR 0.97; P = 0.916) tumours was similar; by contrast, the risk of death associated with T2b (HR 1.81; P = 0.031) and T3 (HR 1.89; P = 0.038) lesions was significantly greater than for T1a tumours. Conclusion: Subclassification of T2 tumours should be undertaken routinely in order to stratify patients with gastric cancer more accurately in terms of their mortality risk.

Macrophage-Mediated Melanoma Reduction after HP-NAP Treatment in a Zebrafish Xenograft Model

Gaia Codolo, Nicola Facchinello, Nicole Papa, Ambra Bertocco, Sara Coletta, Clara Benna, Luigi Dall'Olmo, Simone Mocellin, Natascia Tiso, Marina de Bernard

Summary: The Helicobacter pylori Neutrophil Activating Protein (HP-NAP) possesses immunomodulatory properties and can activate cytotoxic Th1 responses, inhibiting tumor growth and impacting the activity of macrophages. Using a zebrafish model, it was found that HP-NAP efficiently inhibits melanoma growth and metastasis by recruiting macrophages with a pro-inflammatory profile, highlighting the crucial role of activated macrophages in countering tumor growth.

Impact of Wide Local Excision on Melanoma Patient Survival: A Population-Based Study

Summary: By combining clinical and administrative data, this study demonstrates that almost one in ten CMM patients did not undergo wide local excision surgery. Monitoring how diagnostic-therapeutic protocols are actually implemented in the real world may significantly contribute to promoting quality improvements in the management of oncological patients.

Early increase in circulating carbonic anhydrase IX: A potential new predictive biomarker of preeclampsia

Silvia Galbiati, Daniela Gabellini, Alessandro Ambrosi, Nadia Soriani, Federica Pasi, Massimo Locatelli, Roberta Luciano, Massimo Candiani, Luca Valsecchi, Gianpaolo Zerbini, Maddalena Smid

Summary: Preeclampsia is a severe complication of pregnancy, and finding predictive biomarkers is crucial for prevention. This study compared the diagnostic accuracy of carbonic anhydrase IX (CAIX) and sFlt1/PlGF ratio as biomarkers for predicting preeclampsia. CAIX levels significantly increased before the onset of preeclampsia, and it performed better than sFlt1/PlGF ratio in predicting the disease. CAIX could be an effective predictive biomarker for preeclampsia, but further studies are needed to confirm this finding.

Summary: We investigated the correlation between the occurrence of cutaneous melanoma and noncutaneous tumors in CDKN2A mutation carriers and wild-type melanoma patients. The study found that mutation carriers had a significantly higher number of cutaneous melanoma cases compared to the wild-type patients. Additionally, both groups had a similar rate of diagnosis for malignant NCTs. The onset of cutaneous melanoma generally preceded the occurrence of NCTs, emphasizing the importance of comprehensive surveillance for melanoma patients.

Summary: This study aimed to investigate predictors and clinical impact of rebleeding in poor-grade aneurysmal subarachnoid hemorrhage (aSAH). The study found that ultra-early vasospasm (UEV) and presence of dissecting aneurysm were the strongest predictors of aneurysmal rebleeding, while a history of hypertension reduced the chances of rebleeding. Additionally, rebleeding emerged as an independent predictor of intrahospital mortality.

Endometriosis-Related Ovarian Cancers: Evidence for a Dichotomy in the Histogenesis of the Two Associated Histotypes

Alice Bergamini, Giorgia Mangili, Alessandro Ambrosi, Gianluca Taccagni, Emanuela Rabaiotti, Luca Bocciolone, Giorgio Candotti, Raffaella Cioffi, Francesca Pella, Giulia Sabetta, Costanza Saponaro, Massimo Candiani

Summary: The study compares data from patients with endometrioid and clear cell ovarian tumors associated with endometriosis to investigate the hypothesis of a dichotomy in their histogenesis. It found that the occurrence of endometriosis is more frequent in patients with clear cell ovarian cancer. The EAOEC group showed higher bilateral occurrences and a higher solid/cystic rate at gross pathology. ECC patients had a more advanced disease stage and a synchronous endometrial carcinoma was found in 38% of EAEOC patients.

Summary: Vessel wall imaging (VWI) with black-blood (BB) technique can reveal aneurysmal enhancement prior to growth/rupture in treatment-naive cerebral aneurysms. Recent studies suggest that BB enhancement may also occur in endovascularly treated aneurysms, but its significance is debatable. This study explores the relationship between BB enhancement and incomplete occlusion status and coil packing density, hypothesizing a flow-related mechanism.

Summary: Amyotrophic lateral sclerosis (ALS) is a lethal neurodegenerative disease primarily affecting individuals between 50-60 years old. The presence of TDP-43 proteinopathy, characterized by cytoplasmic aggregates formed by TDP-43 protein, disrupts mRNA transport and localization. This study generated and analyzed cellular models of TDP-43 proteinopathy, showing that neurons expressing TDP-43 fusion proteins have impaired axonal protein synthesis, increased oxidative stress, and defects in presynaptic function and electrical activity. These changes are correlated with deregulation of axonal mRNA levels involved in the same processes.

Alessandra Buja, Massimo Rugge, Giuseppe De Luca, Emanuela Bovo, Manuel Zorzi, Chiara De Toni, Claudia Cozzolino, Antonella Vecchiato, Paolo Del Fiore, Romina Spina, Sandro Cinquetti, Vincenzo Baldo, Carlo Riccardo Rossi, Simone Mocellin

Summary: This study focuses on the changing incidence of cutaneous malignant melanoma (CMM) in Veneto region and its Alpine area in Northeast Italy, finding a significant increase in CMM incidence over the past 30 years, particularly among Alpine residents, especially younger females. Clinicopathological profile of CMM is associated with the place of residence. These findings have important implications for CMM prevention strategies.
