Summary: In this study, a enzymatic protocol was developed for the preparation of chemical nuclease-TFO hybrid constructs, which includes the incorporation of novel artificial metallo-nuclease (AMN)-dNTPs into the TFO strand. The specificity of enzymatically synthesised DPA-TFOs was improved through coordinated designer intercalators, and high-precision DNA cleavage was achieved. This research demonstrates the potential of chemical nuclease-TFOs in overcoming the limitations of non-molecularly targeted metallodrugs and advancing artificial gene-editing technology.

Summary: In this study, the coordination properties of ligands were investigated by modifying their side chain structure. The results showed that the length of the aliphatic linker, isomers of the amide group, and the type of chiral terminal group all had an impact on the coordination properties. Additionally, DFT calculations provided insights into the stability of isomers and the potential hydrogen bonding between ligands in a ML2 complex.

Summary: We synthesized and characterized Cu(ii) complexes CuL1-CuL6 with cyclen-based ligands. The ligands L-1-L-6 comprise different donor sets including S and O atoms. The substitution of N donor atoms with hard donor O atoms leads to efficient oxidative nucleases, but dissociation of the complex upon reduction. We are showing that the rational combination of O and S atoms next to two nitrogen donors within the macrocycle leads to the stabilization of both redox states, resulting in the highest oxidative DNA cleavage activity within this family of cyclen-based Cu(ii) complexes.

Summary: Cu(ii) complexes with ATCUN peptide ligands were investigated for their ROS generation and oxidative DNA degradation abilities. However, the biological activity of most ATCUN complexes such as Cu-GGH (Gly-Gly-His) is low. Tuning the redox chemistry by incorporating N-heteroaromatics reinstates ROS production, leading to efficient DNA cleavage.

Summary: Many Schiff bases derived from 2-hydrazinobenzothiazole and isatin have been used to form [ML2]Cl2 type Co(II) complexes. Nitrogen from azomethine, oxygen from lactonyl, and nitrogen atoms of the thiazole ring contribute to coordinating the metallic ion. The complexes consist entirely of metal and have a geometrical octahedron structure, as confirmed by analytical, magnetic, and spectroscopic investigations.

Summary: In this study, four novel Cu complexes with different geometries were synthesized, and their interactions with DNA were investigated. The Cu complexes containing quinoline and pyridine were found to destabilize the DNA structure through minor groove binding and intercalation, respectively. Among them, the Cu2 complex containing quinoline and morpholine exhibited the highest distortion and cleavage efficiency of DNA.

Summary: Copper (II) complexes are potential alternatives to platinum drugs in cancer therapy due to their multi-target mode of action. In this study, new mononuclear and dinuclear coordination compounds of copper were synthesized and characterized. The compounds showed stability in aqueous medium and were found to bind human serum albumin (HSA) with different stoichiometries and affinities. Based on these findings and the known antiproliferative activity of copper complexes, further studies on their potential against tumor cell lines are warranted.
