Summary: The noradrenergic nucleus locus coeruleus plays a crucial role in the brain's response to stress. The neuropeptide corticotropin-releasing factor (CRF) and endogenous opioid peptide enkephalin are both involved in regulating anxiety-like behavior and the behavioral response to stress within the locus coeruleus. The downregulation of delta-opioid receptors (DORs) within the locus coeruleus may mediate the effects of stress on anxiety-like behavior and locus coeruleus activity, while overexpression of DORs can block these effects. Enkephalinergic neurotransmission at DORs within the locus coeruleus is important for the behavioral response to stress and may also play a role in reward-related behavior.

Summary: The search for new drugs for the treatment of acute myocardial infarction and reperfusion injury is urgent. Opioid peptides, such as deltorphin II, have been found to have a cardioprotective effect in reperfusion, and this study aimed to investigate the molecular mechanisms involved. The results showed that the cardioprotective effect of deltorphin II in reperfusion is mediated through the activation of the peripheral delta(2) opioid receptor, most likely located in cardiomyocytes.

Currently available anti-epileptic drugs have significant side effects due to low selectivity to molecular targets. Activation of alpha2B adrenergic receptors (ARs) in the thalamus shows potential as a specific molecular target for absence epilepsy. HCN and calcium channels are proposed as relevant cellular targets of alpha2 ARs in spike-wave activity generation.

Summary: The acute effects of DOR agonist SNC80 and antagonist naltrindole on the firing activity of hippocampal glutamate and brainstem monoamine neurons were investigated. SNC80 dose-dependently inhibited the activity of glutamate, 5-HT, and dopamine neurons, while naltrindole dose-dependently stimulated 5-HT neurons. The co-administration of SNC80 and naltrindole inhibited noradrenaline neurons. These findings suggest that DOR ligands can modulate the firing activity of hippocampal glutamate and brainstem monoamine neurons in vivo conditions.
