The objective of the present study was to assess the effects of Enterococcus sp. strain TN-3 isolated from deep seawater on inhibition of eosinophil accumulation, IgE production and active cutaneous anaphylaxis (ACA). We investigated the effects of viable and non-viable TN-3 on allergen-induced peritoneal eosinophil accumulation in mice. Viable (5.4 X 10(10) colony-forming units per 60 mg) or non-viable TN-3 (60 mg) was orally administered to BALB/c mice that had been sensitised with the cedar pollen (Cryptomeria japonica) allergen. Oral administration of non-viable TN-3 was effective in suppressing eosinophil accumulation while viable TN-3 was ineffective. We also examined the dose-response relationship for non-viable TN-3 in regard to eosinophil accumulation, IgE production and ACA in allergen-primed mice. Non-viable TN-3 was orally administered at doses of 15 mg (low dose), 30 mg (medium dose) and 60 mg (high dose) to BALB/c mice that had been sensitised with cedar pollen allergen. The anti-allergic effects expressed as inhibition of eosinophil accumulation, IgE production and ACA were found at the low and high doses, but not at the medium dose. These results suggest that non-viable TN-3 exhibited anti-allergic effects at doses of 45 and 60 mg.

Comparison of Bioabsorbable Steroid-Eluting Sinus Stents Versus Nasopore After Endoscopic Sinus Surgery: A Multicenter, Randomized, Controlled, Single-Blinded Clinical Trial

Zhenxiao Huang, Bing Zhou, Dehui Wang, Hongrui Zang, Huankang Zhang, Huan Wang, Shenqing Wang, Lei Cheng, Jinrang Li, Wenying Wu, Huifang Zhou, Huili Wu

Summary: This study compared the efficacy of bioabsorbable steroid-eluting sinus stents with absorbable Nasopore packs after endoscopic sinus surgery for chronic rhinosinusitis. Results showed that steroid-eluting stents significantly reduced the need for postoperative surgical intervention and polyp formation compared to Nasopore, and were effective in preventing severe adhesions. Both the stents and Nasopore packs were effective in maintaining ethmoid sinus patency and preventing middle turbinate lateralization. Further prospective cohort studies with long-term outcomes are needed.

Summary: The role of IRF7 in hair cell development in zebrafish was investigated, suggesting it may be a member of the deafness gene family. Knockdown of irf7 led to alterations in the expression of multiple genes, ultimately affecting the development of hair cells.

Summary: This study identified TP53 as the most frequently mutated gene in patients with sarcomatoid carcinoma of the head and neck, with mutations in genes related to cell cycling, chromatin remodeling, and histone modification also being common. Alterations in receptor tyrosine kinases (RTKs) were found in some patients, with PIK3CA being the most frequently mutated gene. The tumor mutation burden (TMB) value was significantly higher in PIK3CA mutation patients compared to wild-type patients.

Summary: Corticosteroids are effective in treating chronic rhinosinusitis, but concerns about potential side effects persist. Transnasal nebulization may offer a balanced solution, yet there is currently no consensus or guidelines on its use. Chinese experts have developed a consensus on the application of steroid transnasal nebulization in treating CRS, focusing on efficacy and safety considerations.

Safety of intranasal corticosteroids for allergic rhinitis in children

Min Zhang, Jing-Zi Ni, Lei Cheng

Summary: Intranasal corticosteroids (INCSs) have a favorable safety profile and high local-systemic balance of bioavailability with a low incidence of adverse events in the treatment of pediatric allergic rhinitis (AR). However, the use of INCSs should be individualized based on patient response and adverse effects. Evaluation of the benefits and risks of INCSs is necessary to ensure clinical efficacy and prevent potential events.

TLR Signaling Pathway Gene Polymorphisms, Gene-Gene and Gene-Environment Interactions in Allergic Rhinitis

Ruo-Xi Chen, Meng-Di Dai, Qing-Zhao Zhang, Mei-Ping Lu, Mei-Lin Wang, Min Yin, Xin-Jie Zhu, Zhong-Fei Wu, Zheng-Dong Zhang, Lei Cheng

Summary: This study explored the genetic association of allergic rhinitis (AR) with single nucleotide polymorphisms (SNPs) in the Toll-like receptor (TLR) signaling pathway. It found that genetic polymorphisms in TLR2 and CD14, as well as gene-gene interactions, were associated with susceptibility to AR in this Han Chinese population. However, the results regarding gene-environment interactions were limited.

Gut microbiome modulation by probiotics, prebiotics, synbiotics and postbiotics: a novel strategy in food allergy prevention and treatment

Jialu Shi, Youfa Wang, Lei Cheng, Jin Wang, Vijaya Raghavan

Summary: Food allergy has become a global public health issue, especially in developed countries. Recent studies have shown that dysbiosis in the gut microbiota is strongly linked to the increasing prevalence of food allergy. Modulating the gut microbial composition is crucial in regulating the immunogenicity of food antigens. Therefore, probiotics, prebiotics, synbiotics, and postbiotics have attracted significant attention as innovative strategies for preventing and treating food allergy.
