BRCA1-associated breast and ovarian cancer risks in Poland: no association with commonly studied polymorphisms

Polymorphisms in genes involved in DNA repair, steroid hormone biosynthesis/metabolism/signaling, folate metabolism as well as cell growth are prime candidates for possible associations with breast and ovarian cancer risk in women with an inherited predisposition. We investigated 29 polymorphisms in 20 genes encoding key proteins of the above four biological pathways for their breast and ovarian cancer risk modifying effect in Polish women harboring BRCA1 founder mutations. Of the analyzed genes, ERCC2, XRCC1, XRCC2, XRCC3 and Lig4 participate in DNA repair, TP53 in cell cycle check point control, AIB1, AR, COMT, CYP11A1, CYP17A1, CYP19A1, HSD17 and PGR in steroid hormone biosynthesis/metabolism/signaling, TYMS in folate metabolism and HER2, IL6, LRP1, TGFB and TGFBR1 affect cell growth. Using validated methods, we genotyped 319 breast cancer cases, 146 ovarian cancer cases and 290 unaffected controls, all of whom harbored one of three causative mutations in BRCA1. Our results revealed no association of any of the investigated polymorphisms with BRCA1-associated breast or ovarian cancer risk. Thus, it appears that these polymorphisms do not influence disease risk in Polish women carrying one of the three common BRCA1 founder mutations.

Summary: Clonal evolution plays a significant role in the progression of CLL. Long-term longitudinal mutation profiling study revealed distinct evolutionary patterns, with minor clonal shifts in stable disease and relapse after long-lasting treatment response, but major clonal shifts in refractory disease. These shifts are not strongly linked to known CLL driver genes, suggesting that they are mostly driven by treatment-induced selection of sub-clones, highlighting the need for novel non-genotoxic treatment regimens.

Summary: CLL is a B-cell malignancy occurring mainly in the elderly, with transgenic expression of TCL1 not sufficient for full leukemic transformation. Studies using E mu-TCL1 mice showed B-cell receptor stereotypy similar to patients, confirming the antigen-driven nature of CLL. Genomic alterations in E mu-TCL1 mouse model mirror human CLL and identify Myc as an oncogenic hit associated with disease progression in patients.

Contribution of large genomic rearrangements in PALB2 to familial breast cancer: implications for genetic testing

Temporal control of the integrated stress response by a stochastic molecular switch

Summary: This study elucidated the molecular mechanism of stress granules formation by integrating quantitative experiments and mathematical modeling. The study revealed that the stress response is controlled by a stochastic switch, with key elements including cooperative activation of PKR, ultrasensitive response of SG formation to eIF2 alpha phosphorylation, and negative feedback via GADD34. Furthermore, the study identified GADD34 mRNA levels as a molecular memory of the ISR that plays a central role in cell adaptation to acute and chronic stress.

Summary: In this prospective study, the associations between three serum elements (Se, Zn, and Cu) and the prognosis of four different types of cancer were measured. Higher levels of selenium and zinc were associated with reduced mortality, while higher levels of copper were associated with increased mortality. These findings suggest that these serum elements may have implications for the prognosis of cancer.

Summary: This study compared the colorectal cancer incidence in carriers of pathogenic variants of the MMR genes in two cohorts. The PLSD cohort, with mandatory colonoscopy surveillance, had higher CRC incidences compared to the retrospective IMRC cohort. Surprisingly, the incidence was not reduced in path_MSH6 carriers. However, the incidence in path_PMS2 carriers was reduced, though not significantly.

Modifier genes and Lynch syndrome: some considerations

Summary: Lynch Syndrome is a highly variable disease with patients presenting different manifestations, from early-onset malignancy to no development of malignancy despite having a genetic predisposition. Colorectal cancer is the most common malignancy associated with LS, involving both genetic and environmental factors. Environmental insults, such as smoking and excessive alcohol consumption, are known to increase cancer risk. However, in individuals with inherited predisposition, the interaction between environmental insults and genetic factors is complex and requires careful consideration.
