Waterham, David Cassiman, Frederic M. Vaz

Summary: The study found that de novo variants affecting the Arg480 residue of FAR1 lead to an autosomal dominant disorder with a different disease mechanism than that of recessive FAR1 deficiency. FAR1 deficiency causes defective ether lipid synthesis, while the de novo variants result in elevated plasmalogen levels. Further functional studies in fibroblasts revealed disruption of plasmalogen-dependent feedback regulation of FAR1 protein levels.

Summary: Unstable hemoglobinopathies (UHs) are rare anemia disorders characterized by abnormal hemoglobin variants with decreased stability, easily causing hemolysis. Diagnosis of UHs requires a high index of suspicion by the treating physician, highlighting the importance of next generation sequencing (NGS) methodologies.

Homozygous UBA5 Variant Leads to Hypomyelination with Thalamic Involvement and Axonal Neuropathy

Murtadha L. Al-Saady, Charlotte S. Kaiser, Felipe Wakasuqui, G. Christoph Korenke, Quinten Waisfisz, Abeltje Polstra, Petra J. W. Pouwels, Marianna Bugiani, Marjo S. van der Knaap, Roelineke J. Lunsing, Eva Liebau, Nicole I. Wolf

Summary: The study found that the enzyme UBA5 plays a crucial role in neurodevelopmental disorders, especially in early-onset epileptic encephalopathy. A homozygous UBA5 variant was detected in a patient with severe global developmental delay and epilepsy, leading to insights into the impact of this variant on UBA5 protein function. The findings suggest that epilepsy associated with UBA5 variants may manifest later in life than previously thought, and radiological signs may include hypomyelination and thalamic involvement.

Fetal akinesia deformation sequence and massive perivillous fibrin deposition resulting in fetal death in six fetuses from one consanguineous couple, including literature review

Summary: This study reported a consanguineous couple with six fetuses developing Fetal Akinesia Deformation Sequence (FADS) and MPFD, possibly due to a genetic cause. The literature review found a low prevalence of FADS and MPFD cases, supporting the conclusion that multiple factors (fetal, fetoplacental, genetic) may contribute to causing MPFD.
