Background: Bombina variegate peptide 8 (Bv8) is a small protein secreted by frog skin. Recently it has been shown to contribute to tumor angiogenesis in mouse model. The purpose of this study was to investigate Bv8 in mice with type II collagen-induced arthritis (CIA). Methods: We induced CIA in male DBA/1J mice. The severity of arthritis was evaluated based on an arthritis score. RNA was extracted from the joint, and examined for Bv8 mRNA expression by RT-PCR and real-time RT-PCR. Synovial tissue and bone marrow were immunohistochemically examined using anti-Bv8 antibody. Results: The level of Bv8 mRNA expression in the joint was below the detection limit in the control group, but was elevated in the CIA group, and was correlated with the arthritis score. In addition, an increase in Bv8-positive cells was observed in the synovium and bone marrow in the CIA group. Conclusion: Bv8 was elevated in the synovium and bone marrow of CIA mice, suggesting that Bv8 plays an important role in the pathogenesis of arthritis.

Differential inflammation-mediated function of prokineticin 2 in the synovial fibroblasts of patients with rheumatoid arthritis compared with osteoarthritis

Summary: The study investigated the role of Prokineticin 2 (PK2) in the pathogenesis of rheumatoid arthritis (RA) and osteoarthritis (OA). PK2 and its receptors were expressed in synovial tissues of both RA and OA patients. The study found that PK2 could suppress the production of IL-6 from OA synovial fibroblasts, but this effect was weaker in RA synovial fibroblasts.

Inhibitory effect of baricitinib on microglia and STAT3 in a region with a weak blood-brain barrier in a mouse model of rheumatoid arthritis

Summary: In patients with rheumatoid arthritis (RA), baricitinib has been shown to improve both arthritis symptoms and neuropsychological symptoms, such as depression and fatigue. This study investigated the effect of baricitinib on microglia in the brain region with a weak blood-brain barrier (BBB) using a CIA mouse model. The results demonstrated that baricitinib decreased activated microglia levels in the early phase and improved food intake and sucrose preference. However, it did not affect microglial morphology in the late phase.

Autocitrullination confers monocyte chemotactic properties to peptidylarginine deiminase 4

Summary: PAD4 contributes to the production of citrullinated proteins as autoantigens in RA. This study showed that autocitrullinated PAD4 can recruit monocytes and induce inflammation in mouse joints. Higher levels of citrullinated PAD4 were found in the synovial fluid of RA patients compared to osteoarthritis patients.

Summary: This article reports a case of developing SLE after SARS-CoV-2 vaccination and suggests that patients who develop SLE within 2 weeks of vaccination are more likely to have immunological abnormalities or a family history of autoimmune diseases. The study also implies the involvement of type I interferon in the pathogenesis of SLE after SARS-CoV-2 vaccination.

How do central sensitisation features affect symptoms among patients with rheumatoid arthritis? Analysis of pain descriptors and the effect of central sensitivity syndrome on patient and evaluator global assessments

Summary: This study investigated central sensitivity syndrome (CSS) in patients with rheumatoid arthritis (RA) and its impact on pain characteristics and evaluator global assessment. The results showed that CSS was associated with evaluator global assessment and PGA minus EGA, and the descriptors of pain differed between disease activity-related pain and CS-related pain in RA patients.

Central sensitisation features are associated with neuropathic pain-like symptoms in patients with longstanding rheumatoid arthritis: a cross-sectional study using the central sensitisation inventory

Summary: This study aims to explore the prevalence, clinical characteristics, and association between central sensitisation syndrome (CSS) and neuropathic pain (NP)-like symptoms in rheumatoid arthritis (RA) patients. The findings suggest that NP-like symptoms might be one of the symptoms of CSS in longstanding RA patients. Detecting CSS using the central sensitisation inventory (CSI) may help to understand the pathogenesis of patients with disproportionately greater NP-like symptoms and/or widespread pain compared with the degree of inflammation.
