Fonsecaea pugnacius, a Novel Agent of Disseminated Chromoblastomycosis

We report a fatal case of a chromoblastomycosis-like infection caused by a novel species of Fonsecaea in a 52-year-old immunocompetent Caucasian male from an area of chromoblastomycosis endemicity in Brazil. The patient had a 30-year history of slowly evolving, verrucous lesions on the right upper arm which gradually affected the entire arm, the left hemifacial area, and the nose. Subsequent dissemination to the brain was observed, which led to death of the patient. The internal transcribed spacer (ITS) and partial large subunit (LSU), BT2, and CDC42 genes of the isolates recovered from skin and brain were sequenced, confirming the novelty of the species. The species is clinically unique in causing brain abscesses secondary to chromoblastomycosis lesions despite the apparent intact immunity of the patient. Histopathologic appearances were very different, showing muriform cells in skin and hyphae in brain.

Summary: This case report describes a patient with fungal keratitis caused by Roussoella neopustulans. The patient experienced visual improvement after topical natamycin treatment and regained sight after three months of therapy. This is the first reported case of keratitis associated with R. neopustulans.

Sporotrichosis in Children: Case series and Narrative Review

Summary: This review evaluates the risk and prognostic factors related to pediatric sporotrichosis. It found that Sporothrix schenckii is the major etiologic agent in Mexico, transmitted sapronotically, while Sporothrix brasiliensis is mainly transmitted by cats in Brazil. The clinical manifestations and affected anatomical sites differ between the two countries.

Summary: This study demonstrated for the first time that different species of Fonsecaea produce extracellular vesicles (EVs) in response to nutritional conditions, and these EVs can modulate the production of pro- and anti-inflammatory cytokines and nitric oxide by bone marrow-derived macrophages (BMDMs). The size, content, and morphology of the EVs were also affected by growth conditions.

Summary: This study investigated the presence of black fungi in the skin microbiome and found that Exophiala and Cyphellophora europaea were the most common species. Summary: This study focused on the sequencing, assembly, and description of the whole genome of Cladophialophora exuberans, a black yeast related fungus, with an emphasis on genes and pathways related to carbon and toxin management, tolerance and bioremediation of lead and copper, and metal homeostasis. The results showed that the strain exhibited growth inhibition at minimum inhibitory and fungicidal concentrations of copper and lead, and could grow at higher concentrations in agar tests. In addition, the strain demonstrated uptake capacities of copper and lead in heavy metal bioremediation assays. This study contributed to the annotation of genes involved in heavy metal homeostasis and provided insights into the mechanisms of tolerance and adaptation to extreme conditions.

Summary: Fungi in the Chaetothyriales order, although not primary pathogens, have the ability to cause human infections. They exhibit extremotolerance and advanced toxin management, and their ancestral ecology involves association with metabolite-producing lichens. Ant-association and dealing with pheromones are also important aspects of this order. The Herpotrichiellaceae family, derived from phylogeny, shows dual ecology and the ability to cause disease in humans and cold-blooded vertebrates.

Genotyping and antifungal susceptibility testing of Sporothrix brasiliensis isolates from Southern Brazil

Summary: This study analyzed the genetic relatedness and antifungal susceptibility of 89 Sporothrix isolates from humans and cats in Curitiba, Southern Brazil. The results showed zoonotic transmission and a broad activity of seven common antifungals, including olorofim, against a large S. brasiliensis isolate collection. Only one human isolate exhibited resistance to itraconazole and posaconazole. Whole genome sequencing analysis did not reveal any unique substitutions in resistance-associated genes.

Nannizzia species causing dermatophytosis in cats and dogs: First report of Nannizzia incurvata as an etiological agent in Brazil

Summary: This study aimed to evaluate the current prevalence of Nannizzia-causing canine and feline dermatophytosis in Curitiba and Metropolitan Region, Brazil, as well as perform phenotypic and phylogenetic characterizations of these isolates. The results showed that dogs were significantly more affected by Nannizzia than cats in terms of dermatophytes infection. Furthermore, molecular analysis confirmed the first report of N. incurvata as an etiological agent of canine and feline dermatophytosis in Brazil.
