Glucosidase inhibitory activity and antioxidant activity of flavonoid compound and triterpenoid compound from Agrimonia Pilosa Ledeb

Background: In Chinese traditional medicine, Agrimonia pilosa Ledeb (APL) exhibits great effect on treatment of type 2 diabetes mellitus (T2DM), however its mechanism is still unknown. Considering that T2DM are correlated with postprandial hyperglycemia and oxidative stress, we investigated the alpha-glucosidase inhibitory activity and the antioxidant activity of flavonoid compound (FC) and triterpenoid compound (TC) from APL. Methods: Entire plants of APL were extracted using 95% ethanol and 50% ethanol successively. The resulting extracts were partitioned and isolated by applying liquid chromatography using silica gel column and Sephadex LH 20 column to give FC and TC. The content of total flavonoids in FC and the content of total triterpenoids in TC were determined by using UV spectrophotometry. HPLC analysis was used to identify and quantify the monomeric compound in FC and TC. The alpha-glucosidase inhibitory activities were determined using the chromogenic method with p-nitrophenyl-alpha-D-glucopyranoside as substrate. Antioxidant activities were assessed through three kinds of radical scavenging assays (DPPH radical, ABTS radical and hydroxyl radical) & beta-carotene-linoleic acid assay. Results: The results indicate FC is abundant of quercitrin, and hyperoside, and TC is abundant of 1 beta, 2 beta, 3 beta, 19 alpha-tetrahydroxy-12-en-28-oic acid (265.2 mg/g) and corosolic acid (100.9 mg/g). The FC & the TC have strong alpha-glucosidase inhibitory activities with IC50 of 8.72 mu g/mL and 3.67 mu g/mL, respectively. We find that FC show competitive inhibition against alpha-glucosidase, while the TC exhibits noncompetitive inhibition. Furthermore, The FC exhibits significant radical scavenging activity with the EC50 values of 7.73 mu g/mL, 3.64 mu g/mL and 5.90 mu g/mL on DPPH radical, hydroxyl radical and ABTS radical, respectively. The FC also shows moderate anti-lipid peroxidation activity with the IC50 values of 41.77 mu g/mL on inhibiting beta-carotene bleaching. Conclusion: These results imply that the FC and the TC could be responsible for the good clinical effects of APL on T2MD through targeting oxidative stress and postprandial hyperglycaemia. So APL may be good sources of natural antioxidants and alpha-glucosidase inhibitors exhibiting remarkable potential value for the therapy of T2DM.

Summary: This study investigated the microbial diversity and functional prediction of rhizosphere microbes associated with wild and cultivated Glycyrrhiza uralensis Fisch plants. The results showed that the bacterial communities in wild samples were dominated by Proteobacteria, while Actinobacteria dominated the cultivated samples. The observed species number was higher in cultivated soil samples, and the weighted-unifrac distance of two cultivated samples was closer despite being from different regions.

Synergistic Plant-Microbe Interactions between Endophytic Actinobacteria and Their Role in Plant Growth Promotion and Biological Control of Cotton under Salt Stress

Summary: In this study, 71 bacterial endophytes associated with the medicinal plant Thymus roseus were screened for their plant growth promotion (PGP) abilities and biocontrol potential. The results showed that certain strains had positive effects on plant growth, pathogen resistance, and stress tolerance. Strains XIEG05 and XIEG12 significantly enhanced plant tolerance to salt stress, while strain XIEG57 reduced cotton disease incidence caused by Verticillium dahliae. GC-MS analysis revealed the presence of antimicrobial and antifungal compounds in these strains. These findings provide valuable insights for the application of these strains as bioinoculant fertilizers and biological control agents in saline soil conditions.

Bacterial Community Structure and Potential Microbial Coexistence Mechanism Associated with Three Halophytes Adapting to the Extremely Hypersaline Environment

Summary: This study investigated the diversity and community structure of endophytic and rhizospheric bacteria associated with three halophytes. The results showed that rhizospheric bacterial richness and diversity were higher than for endophytic bacteria. Actinobacteria, Firmicutes, Bacteroidetes and Proteobacteria were the dominant bacterial phyla. This research provides important insights into the ecological adaptation mechanism of halophytes.

Sediment prokaryotic microbial community and potential biogeochemical cycle from saline lakes shaped by habitat

Summary: This study investigated the distribution and potential functions of prokaryotes in different saline lakes in Xinjiang using the cultivation-independent method of amplicon sequencing. The results showed that Proteobacteria was the predominant community in all types of saline lakes, while Desulfobacterota, Firmicutes, Acidobacteriota, and Chloroflexi were mainly distributed in hypersaline, arid saline, and light saltwater lakes respectively. The functional group analysis revealed that fermentation was the main metabolic process of microbes in all saline lakes. Environmental factors such as SO42-, Na+, CO32-, and TN were found to significantly affect the microbial community.
