Summary: In this study, we observed an upregulation of BIRC5 in cisplatin-resistant cell lines, which was associated with enhanced chemoresistance. Intriguingly, BIRC5 exhibited a high number of m(6)A binding sites. Modification of m(6)A was found to enhance BIRC5 expression. IGF2BP1 stabilized BIRC5 mRNA and synergized with METTL3 to intensify chemoresistance. Our in vivo experiments showed that tumors were significantly smaller in size and volume when BIRC5 was silenced. These results underscore the importance of BIRC5 in chemoresistance.

Summary: This study found that HJURP is overexpressed in ovarian cancer and is associated with unfavorable prognosis. Knockdown of HJURP inhibits proliferation, metastasis, and induces cell stagnation in ovarian cancer cells. Mechanistically, HJURP regulates WEE1 through the MYC pathway, affecting chemosensitivity and DNA repair in ovarian cancer cells. Moreover, silencing HJURP enhances sensitivity to cisplatin and improves the synergistic effect of combined therapy with cisplatin and AZD1775 in ovarian cancer cells.

Summary: Ovarian cancer (OC) has a poor prognosis and resistance to cisplatin (CDDP) treatment due to high expression of ATP-binding cassette (ABC) transporters. This study investigated the role of sortilin-related receptor 1 (SORL1) in OC response to CDDP. SORL1 was found to be closely related to CDDP resistance and predicts a poor prognosis in OC. Knockdown of SORL1 enhanced the effect of CDDP on resistant OC cells by inhibiting the EEA1 pathway and sensitizing cells to CDDP.

Summary: Ovarian cancer is the second most common gynecological malignancy with poor survival rates. Chemoresistance, a major obstacle in treating ovarian cancer, is influenced by tumor microenvironment and soluble factors. FDA-approved JAK inhibitors can partially restore platinum sensitivity in ovarian cancer cells in the tumor microenvironment. Crizotinib, in combination with platinum, can block the promotion of platinum resistance by HGF and restore platinum sensitivity in ovarian cancer cells. Additionally, 2HE2 can restore platinum sensitivity in ovarian cancer cells exposed to the tumor microenvironment.
