Apoptosis inhibitor-5 overexpression is associated with tumor progression and poor prognosis in patients with cervical cancer

Background: The apoptosis inhibitor-5 (API5), anti-apoptosis protein, is considered a key molecule in the tumor progression and malignant phenotype of tumor cells. Here, we investigated API5 expression in cervical cancer, its clinical significance, and its relationship with phosphorylated extracellular signal-regulated kinase 1 and 2 (pERK1/2) in development and progression of cervical cancer. Methods: API5 effects on cell growth were assessed in cervical cancer cell lines. API5 and pERK1/2 immunohistochemical staining were performed on a cervical cancer tissue microarray consisting of 173 primary cervical cancers, 306 cervical intraepithelial neoplasias (CINs), and 429 matched normal tissues. Results: API5 overexpression promoted cell proliferation and colony formation in CaSki cells, whereas API5 knockdown inhibited the both properties in HeLa cells. Immunohistochemical staining showed that API5 expression increased during the normal to tumor transition of cervical carcinoma (P < 0.001), and this increased expression was significantly associated with tumor stage (P = 0.004), tumor grade (P < 0.001), and chemo-radiation response (P = 0.004). API5 expression levels were positively associated with pERK1/2 in cervical cancer (P < 0.001) and high grade CIN (P = 0.031). In multivariate analysis, API5+ (P = 0.039) and combined API5+/pERK1/2+ (P = 0.032) were independent prognostic factors for overall survival. Conclusions: API5 expression is associated with pERK1/2 in a subset of cervical cancer patients and its expression predicts poor overall survival, supporting that API5 may be a promising novel target for therapeutic interventions.

High-Dose Selenium Induces Ferroptotic Cell Death in Ovarian Cancer

Summary: This study found that high-dose sodium selenite (SS) can inhibit the proliferation and induce cell death of ovarian cancer cells through increased reactive oxygen species (ROS) generation. High-dose SS also reduced the levels of glutathione peroxidase (GPx) and triggered lipid peroxidation and ferroptosis via dysregulated GPx4 pathways. In mouse models, high-dose SS significantly reduced tumor growth and weight, and decreased GPx4 expression in tumors. Therefore, high-dose SS is considered a potential chemotherapeutic agent for ovarian cancer, utilizing ferroptotic therapy by depleting glutathione and increasing ROS generation.

The Application of Guanidinium to Improve Biomolecule Quality in Fixed, Paraffin-embedded Tissue

Summary: Neutral buffered formalin (NBF) is commonly used as a fixative in clinical applications, but it damages proteins and nucleic acids, limiting the quality of molecular analysis. Previous studies have shown that BE70, a fixative of buffered 70% ethanol, has advantages over NBF, but protein and nucleic acid degradation in archival paraffin blocks is still a challenge. With the addition of guanidinium salts, BE70G fixative improves the quality of molecular analysis, provides better protein and RNA quality at shorter fixation times, and reduces protein degradation in archival tissue blocks.

High ATP6V1B1 expression is associated with poor prognosis and platinum-based chemotherapy resistance in epithelial ovarian cancer

Summary: The study aimed to investigate the function, molecular mechanisms, and clinical significance of ATP6V1B1 in epithelial ovarian cancer (EOC). It was found that ATP6V1B1 was highly expressed in EOC tissues and was associated with clinicopathological features and prognosis. Knockdown of ATP6V1B1 inhibited cancer cell proliferation and colony-forming abilities. Therefore, ATP6V1B1 could serve as a biomarker for prognostic evaluation and chemotherapy resistance, as well as a potential therapeutic target in EOC.

TRPV1 inhibition overcomes cisplatin resistance by blocking autophagy-mediated hyperactivation of EGFR signaling pathway

Summary: The emergence of cisplatin resistance and side-effects are important factors in cancer treatment failure. The authors identify autophagy-mediated EGFR hyperactivation by TRPV1 promotion as a mechanism of cisplatin resistance and demonstrate the efficacy of a TRPV1 inhibitor to sensitize lung cancer to cisplatin. Future studies should provide evidence regarding the mechanisms of potential targets that can overcome the resistance as well as alleviate pain.

Stratifying the risk of ovarian cancer incidence by histologic subtypes in the Korean Epithelial Ovarian Cancer Study (Ko-EVE)

Summary: This study aimed to verify the association between ovarian cancer (OC) and reproductive- and lifestyle-related risk factors stratified by the subtype of OC. The results showed that smoking was positively associated with high-grade serous (HGS) OC, alcohol consumption was positively associated with mucinous type (MUC), and obesity was associated with clear cell type (CLC). Further, spontaneous abortion was negatively associated with CLC, while tubal ligation, hysterectomy, and oophorectomy were associated with decreased risk of HGS. Early menarche was strongly associated with increased risk of CLC. Additionally, childbirth, oral contraceptives, and hormone replacement therapy were negatively associated with OC.

Nectin-4 as a Predictive Marker for Poor Prognosis of Endometrial Cancer with Mismatch Repair Impairment

Prognostic value of β-Arrestins in combination with glucocorticoid receptor in epithelial ovarian cancer

Summary: Hormonal factors, particularly steroid hormones, are believed to play a significant role in ovarian cancer development. The expression of glucocorticoid receptor (GR) has been associated with poor prognosis in epithelial ovarian cancer. This study investigates the clinical significance of beta-arrestin expression in ovarian cancer and the potential synergistic effect with GR as prognostic factors. The findings demonstrate a moderate correlation between beta-arrestin 1 and GR expression, suggesting a possible role in prognostication.

Selective ablation of TRA-1-60+pluripotent stem cells suppresses tumor growth of prostate cancer

Summary: This study investigated the clinical implications of TRA expression in prostate cancer. It found that high PODXL copy number alterations were associated with poor prognosis. TRA-targeted PET imaging successfully located cancer stem cells (CSCs) and TRA-targeted RPT showed promising therapeutic effects. Ablation of TRA+ CSCs inhibited prostate cancer growth.

East Asian Gynecologic Oncology Trial Group (EAGOT): founding history and future perspective

Summary: There are racial and regional differences in gynecologic cancer, but most large-scale phase III studies have been conducted in Western countries, limiting data on Asians. To address this, four clinical trial groups in East Asia have established the East Asia Gynecologic Oncology Trial Group (EAGOT) to conduct international clinical trials and provide the best treatments for Asian women. EAGOT aims to establish standards of care and guidelines for gynecologic cancer in Asia.
