Induction of protective and therapeutic anti-pancreatic cancer immunity using a reconstructed MUC1 DNA vaccine

Background: Pancreatic cancer is a common, highly lethal disease with a rising incidence. MUC1 is a tumor-associated antigen that is over-expressed in pancreatic adenocarcinoma. Active immunotherapy that targets MUC1 could have great treatment value. Here we investigated the preventive and therapeutic effect of a MUC1 DNA vaccine on the pancreatic cancer. Methods: MUC1-various tandem repeat units(VNTR) DNA vaccine was produced by cloning one repeat of VNTR and inserting the cloned gene into the pcDNA3.1. In the preventive group, female C57BL/6 mice were immunized with the vaccine, pcDNA3.1 or PBS; and challenged with panc02-MUC1 or panc02 cell. In the therapeutic group the mice were challenged with panc02-MUC1 or panc02 cell, and then immunized with the vaccine, pcDNA3.1 or PBS. The tumor size and the survival time of the animals were compared between these groups. Results: The DNA vaccine pcDNA3.1-VNTR could raise cytotoxic T lymphocyte (CTL) activity specific for MUC1. In the preventive experiment, the mice survival time was significantly longer in the vaccine group than in the control groups (P < 0.05). In the therapeutic experiment, the DNA vaccine prolonged the survival time of the panc02-MUC1-bearing mice (P < 0.05). In both the preventive and therapeutic experiments, the tumor size was significantly less in the vaccine group than in the control groups (P < 0.05). This pcDNA3.1-VNTR vaccine, however, could not prevent the mice attacked by panc02 cells and had no therapeutic effect on the mice attacked by panc02 cells. Conclusion: The MUC1 DNA vaccine pcDNA3.1-VNTR could induce a significant MUC1-specific CTL response; and had both prophylactic and therapeutic effect on panc02-MUC1 tumors. This vaccine might be used as a new adjuvant strategy against pancreatic cancer.

Significant Enhancement of the Upconversion Emission in Highly Er3+-Doped Nanoparticles at Cryogenic Temperatures

Summary: Relatively low efficiency is the bottleneck for the application of lanthanide-doped upconversion nanoparticles. The high-level doping strategy realized in recent years has not improved the efficiency as much as expected. This study combines theoretical simulation and spectroscopy to elucidate the role of cross relaxation (CR) in the upconversion process and demonstrates that inhibiting CR significantly improves upconversion efficiency.

Responsive Magnetic Nanocomposites for Intelligent Shape-Morphing Microrobots

Summary: With advanced biomedical theragnosis and bioengineering tools, smart and soft responsive microstructures and nanostructures have been developed that can change shapes and convert external power into mechanical actions. This article surveys the progress in designing responsive polymer-particle nanocomposites, leading to the development of smart shapemorphing microscale robotic devices. The use of magnetic fields for tether-free control enables penetration of biological tissues, and future fabrication techniques will be crucial for integrating nanoscale material functionalities into microscale robots.

Metasurface-Controlled Photonic Rashba Effect for Upconversion Photoluminescence

Summary: We demonstrate the effect of spin-momentum locking on upconversion photoluminescence emitted from rare-earth doped nanocrystals coupled to a phase-gradient dielectric metasurface. Different directionalities for left and right circular polarized light are observed, which can be attributed to the photonic Rashba effect that leads to a spin-dependent splitting of emitted light in momentum space.

On-Chip Mirror Enhanced Multiphoton Upconversion Super-Resolution Microscopy

Summary: Multiphoton upconversion super-resolution microscopy (MPUM) is a promising imaging modality that achieves increased resolution and penetration depth through the use of nonlinear near-infrared emission light. However, high excitation power is required for emission saturation, leading to increased phototoxicity. In this study, the authors propose a simple on-chip mirror design that allows for nonlinear saturation emission under low excitation power. This method significantly decreases the excitation power and enables a lateral resolution of around 35 nm in imaging single nanoparticles on-chip. The findings offer a straightforward solution for enhancing super-resolution imaging using pre-designed on-chip devices.

Field-dependent deep learning enables high-throughput whole-cell 3D super-resolution imaging

Summary: We propose a deep-learning method for precise localization of spatially variant point emitters, covering the full chip of a modern sCMOS camera. By using a graphic processing unit-based vectorial point spread function fitter, we accurately model the spatially variant PSF of a high numerical aperture objective in the entire field of view. Combined with deformable mirror-based optimal PSF engineering, we demonstrate high-accuracy three-dimensional single-molecule localization microscopy over a large volume, enabling imaging of mitochondria and nuclear pore complexes in entire cells without hardware scanning, with a 100-fold increase in throughput compared to existing techniques.

Summary: Researchers have developed a modular hydrogel membrane carrier with composite DNAzymes for rapid and highly sensitive detection of pathogenic bacteria gene target analytes. By incorporating composite DNAzymes into a polymer network, the researchers were able to release the DNAzymes to cleave RNA reporter probes and generate fluorescence, achieving quantitative assay performance. The work represents a significant advancement in the development of a versatile pathogenic bacteria testing platform. Overall, the importance of this research is 9 out of 10.

Summary: Researchers have developed a high-resolution near-infrared-II (NIR-II) imaging method for non-invasive diagnosis of skeletal disorders. By using lanthanide-doped nanocrystals, they were able to passively target the bone marrow and achieve effective bone imaging for over two months. The NIR-II imaging technique showed comparable results to X-ray imaging and could also diagnose early onset inflammation in rheumatoid arthritis.

Fourier Channel Attention Powered Lightweight Network for Image Segmentation

Summary: This paper introduces a lightweight network FRUNet based on U-Net, which combines the advantages of Fourier Channel Attention (FCA Block) and Residual unit to improve image segmentation accuracy. FCA Block automatically assigns the weight of learned frequency information to the spatial domain, paying more attention to the precise high-frequency information of diverse biomedical images. The combination of FCA and U-Net is studied, with the skip connection of U-Net fusing encoder information with the decoder. Extensive experimental results show that FRUNet outperforms other advanced medical image segmentation methods in terms of network parameters and accuracy, particularly excelling in pathological section segmentation of nuclei and glands.

A single-cell based mass cytometry study on heterogeneous interactions between upconversion nanoparticles and human immune cells

Summary: In order to promote the use of lanthanide-doped upconversion nanoparticles (UCNPs) in various biomedical applications, it is important to assess their toxicity comprehensively, including their cellular association and interactions with human immune cells. Recent advancements in high-throughput single-cell technologies have enabled us to gain a better understanding of the complex interactions between nanoparticles and human immune cells. In this study, we used a single-cell based mass cytometry technique to investigate the heterogeneous interactions of Yb3+/Er3+-doped NaYF4 UCNPs with human peripheral blood mononuclear cells (hPBMCs). The study demonstrated a new innovative approach to understanding the heterogeneous interactions of conventional nanomaterials with complex human immune systems, which will facilitate the safe implementation of nanomaterials in biomedical applications.
