AMDORAP: Non-targeted metabolic profiling based on high-resolution LC-MS

Background: Liquid chromatography-mass spectrometry (LC-MS) utilizing the high-resolution power of an orbitrap is an important analytical technique for both metabolomics and proteomics. Most important feature of the orbitrap is excellent mass accuracy. Thus, it is necessary to convert raw data to accurate and reliable m/z values for metabolic fingerprinting by high-resolution LC-MS. Results: In the present study, we developed a novel, easy-to-use and straightforward m/z detection method, AMDORAP. For assessing the performance, we used real biological samples, Bacillus subtilis strains 168 and MGB874, in the positive mode by LC-orbitrap. For 14 identified compounds by measuring the authentic compounds, we compared obtained m/z values with other LC-MS processing tools. The errors by AMDORAP were distributed within +/- 3 ppm and showed the best performance in m/z value accuracy. Conclusions: Our method can detect m/z values of biological samples much more accurately than other LC-MS analysis tools. AMDORAP allows us to address the relationships between biological effects and cellular metabolites based on accurate m/z values. Obtaining the accurate m/z values from raw data should be indispensable as a starting point for comparative LC-orbitrap analysis. Summary: Azole resistance in Aspergillus fumigatus is mainly achieved through mutations in the Cyp51A gene and tandem repeat acquisition in the promoter region. However, this study identified other gene mutations such as MfsD, AgcA, UbcD, AbcJ, and RttA contributing to decreased susceptibility to azoles. These findings suggest that various genetic mutations play a role in reducing susceptibility to medical and agricultural azole fungicides.

Intimate genetic relationships and fungicide resistance in multiple strains of Aspergillus fumigatus isolated from a plant bulb

Dielectrophoresis-Based Selective Droplet Extraction Microfluidic Device for Single-Cell Analysis

Summary: We developed a microfluidic device that utilizes dielectrophoresis to selectively trap and extract droplets from multiple trapping pockets. The device includes a main microchannel, five droplet-trapping pockets with side channels, and drive electrode pairs positioned around the trapping pockets. Agarose droplets containing biological samples were successfully trapped in the pockets due to flow resistance differences. Under an applied voltage of 500 V, target droplets were selectively extracted using dielectrophoretic force. The device allows for the screening of encapsulated single cells and the targeted extraction of desired droplets.

Mechanism of Polyhexamethylene Biguanide Resistance in Purpureocillium lilacinum Strains

Summary: This study explores the resistance mechanism of Purpureocillium lilacinum to polyhexamethylene biguanide hydrochloride (PHMB). Genetic mutations were found in PHMB-resistant strains of P. lilacinum, and the knockout of the mutant gene resulted in reduced resistance to PHMB. The adenosine deaminase gene (ADA) was identified as a potential important resistance factor in PHMB-resistant P. lilacinum.
