Nanometer- and angstrom-scale characteristics that modulate complement responses to nanoparticles

Summary: The contribution of the complement system to host defence and homeostasis is well recognized. The mechanisms underlying complement activation by particulate systems are still not fully understood. Activation of the complement cascade can promote inflammatory reactions. Controlled antibody binding and sensing of particles by complement pattern-recognition molecules can trigger complement activation. Particle curvature and spacing arrangement of surface functional groups are important parameters modulating complement responses through engagement with antibodies and complement pattern-recognition molecules. Better understanding of these parameters could enable the engineering of drug carriers and biomedical platforms with tunable complement responses.

Decreased mannan-binding lectin level in adults with hypopituitarism; dependence on appropriate hormone replacement therapies

Summary: Hypopituitarism in adults is associated with a decrease in blood concentration of mannan-binding lectin, which does not occur in hypopituitary patients on appropriate hormone replacement therapies. Therefore, measuring mannan-binding lectin levels may contribute to optimizing hormone replacement therapy doses.

Plasma-derived and recombinant C1 esterase inhibitor: Binding profiles and neuroprotective properties in brain ischemia/reperfusion injury

Summary: The study demonstrates that rhC1INH shows stronger neuroprotective effects after experimental ischemia/reperfusion injury in the brain, reducing behavioral deficits and neuronal loss, decreasing MBL deposition in the brain, and reducing the formation of complement C4b active fragments. In contrast, pdC1INH, despite having a longer plasma residence time, does not exhibit these neuroprotective effects. Therefore, rhC1INH may be a promising drug for stroke as it effectively inhibits MBL.

The disease is caused by the protozoan Leishmania (Leishmania) infantum, transmitted by infected sandflies. Research shows that collectins and ficolins may play a role in the initial immune response to Leishmania infantum by activating the complement system.

Summary: Brain fog, a constellation of neurological and psychiatric symptoms, can occur in the post-acute phase of COVID-19. It is characterized by inattention, short-term memory loss, and reduced mental acuity, which can impact cognition, concentration, and sleep. This study found that levels of Mannan-binding lectin (MBL), an important component of the complement system, were significantly lower in COVID-19 patients experiencing brain fog compared to recovered patients. These findings suggest that MBL deficiency may contribute to the development of long COVID-associated brain fog.

Erysipelothrix rhusiopathiae-specific T-cell responses after experimental infection of chickens selectively bred for high and low serum levels of mannose-binding lectin

Summary: This study investigated immune responses to Erysipelothrix rhusiopathiae (ER) infection in chickens bred for high and low serum mannose-binding lectin (MBL) levels. The results showed that the immune response to ER infection varied, and there were no clear differences between chickens with high and low MBL levels. However, antigen-specific T-cell blast transformation was observed in all infected chickens, and the proportions of blast transformed cells were significantly higher in chickens with low MBL levels. This is the first observation of ER-specific T-cells in chickens, indicating a Th1-type immune response.

The relationship between COVID-19 and the complement system: mannose-binding lectin

Summary: The study found that decreased complement levels before pregnancy are associated with an increased risk of adverse pregnancy outcomes in women with triple antiphospholipid antibodies positivity, while this trend was not observed in those with single or double positivity. Complement levels are cost-effective and easy to measure, making them a potentially useful tool for identifying pregnant women at risk of pregnancy loss.

Antibodies Recognizing Yersinia enterocolitica Lipopolysaccharides of Various Chemotypes in Synovial Fluids From Patients With Juvenile Idiopathic Arthritis

Summary: This study found that chemotype-specific antibodies to Yersinia enterocolitica O:3 LPS were present in synovial fluids from patients with arthritis, with high titers of antibodies specifically recognizing the Kdo-lipid A region. Western blot analysis showed antibodies reacting with fast-migrating LPS fractions and enterobacterial common antigen (ECA). Furthermore, in vitro experiments confirmed that Yersinia LPS processing leads to the exposure of its core region and enhanced potency of complement lectin pathway activation.
