However, there are concerns about the safety of TYK2 inhibition due to the increased risk of lung cancer and non-Hodgkin lymphoma associated with similar medications. Genetic analysis using a loss-of-function variant in TYK2 showed an increased risk of lung cancer and non-Hodgkin lymphoma with therapeutic TYK2 inhibition. These findings have important implications for the safety assessment of deucravacitinib and other TYK2 inhibitors.

The study found that Eastern Asia, Northern America, and South-Central Asia had the highest number of NHL cases. Incidence rates were higher in men than in women, with the highest rates in Australia, New Zealand, Northern America, Northern Europe, and Western Europe. The highest mortality rates were found in regions in Africa, Western Asia, and Oceania.

The presence of autoantibodies was observed in 30 out of 39 patients, while 9 patients did not have autoantibodies. However, there were no statistically significant differences between the two groups in terms of clinical, demographic, staging, and prognosis characteristics. Further research on a broader panel of autoantibodies is needed to explore the relationship between their presence and the clinical outcome of patients.

Summary: Age-related comorbid conditions, such as type 2 diabetes, are common in patients with chronic lymphocytic leukemia (CLL). This study analyzed two separate cohorts to assess the impact of type 2 diabetes on CLL patient outcomes. The results showed that CLL patients with type 2 diabetes had shorter overall survival, were less likely to receive treatment, and had an increased risk of death due to infections. These findings highlight the need for additional interventions and research to address the treatment needs of CLL patients with co-occurring type 2 diabetes.

Summary: We developed a deep learning framework for polygenic risk score (PRS) prediction that can handle large-scale genomics data, support multi-task learning, and automatically integrate clinical and biochemical data. The framework demonstrated competitive performance and improved predictions for complex genetic relationships and non-additive genetic effects and epistasis. The model also outperformed traditional linear PRS methods for Type 1 Diabetes.

Summary: A nationwide COVID-19 lockdown in Denmark resulted in a reduced prevalence of extremely preterm or extremely low birthweight births. This study aimed to investigate the impact of the lockdown on term birthweights in Denmark. The findings showed a small but significant increase in birthweight during the lockdown period, primarily driven by an increase in birthweight in gestational weeks 40 and 41. Additionally, the proportion of infants with a large-for-gestational-age (LGA) increased during the lockdown.
