This review explores the grey area between these two positions and provides a novel analysis of structuralism, highlighting its motivations and conflicts with adaptationist approaches. It clarifies the value of the evo-devo approach to phenotypic diversity and its relationship to other approaches in evolutionary biology.

Summary: The genomic, epigenetic, and metabolic determinants of prostate cancer have been extensively studied in epithelial cells. However, the importance and dynamic role of stromal cells, including fibroblasts, smooth muscle cells, and vasculature-associated cells, in the tumor microenvironment (TME) have been largely overlooked. This review discusses the role of these mesenchymal cell populations in the transition from healthy prostate epithelium to cancer, highlighting their potential as biomarkers and therapeutic targets for prostate cancer.

Summary: Metformin has been widely used to treat diabetes mellitus due to its safety profile and low cost. However, its pharmacokinetics vary among patients, and its poor oral absorption leads to high therapeutic doses and unpleasant gastrointestinal side effects. Novel derivatives of metformin have been synthesized, especially after the identification of organic cation transporters as its main carriers. These derivatives have shown potential in the treatment of various diseases, including cancers and neurodegenerative diseases.

Summary: Toll-like receptor 9 (TLR9) is an intracellular innate immunity receptor that plays a crucial role in chronic inflammation and recognizes pathogenic and self-DNA. Activation of TLR9 leads to the transcription of immune-related or malignancy genes through specific transcription factors. This article reviews the roles of TLR9 in glioma pathogenesis and its related signaling molecules in defending or promoting glioma. TLR9 mediates invasion-induced hypoxia in brain cancer cells by activating matrix metalloproteinases. On the other hand, the TLR9 agonist CpG ODN enhances the antitumor effects of T cells when combined with radiotherapy.

Summary: The imbalance between reactive oxygen species (ROS) production and clearance causes oxidative stress and plays a central role in regulating cell and tissue physiology and pathology. ROS can either stimulate cancer survival and growth or cause cell death depending on their concentration. The transcription factor NRF2 has dual roles in carcinogenesis, where short-term activation suppresses tissue injury, inflammation, and cancer initiation, while constitutive activation confers advantages for cancer cells' survival and growth. NRF2 hyperactivation, as an adaptive cancer phenotype, regulates multiple aspects of cancer development, metastasis, and resistance to anticancer therapy.

Further research based on extrapolating bioactivities to other DAP sub-classes could lead to the development of new therapeutic chemicals. Further evaluation of high-dose toxicity of urolithins and the potential of alternariol derivatives for therapeutic development is recommended.
