Determination of the cardiac drug amiodarone and its N-desethyl metabolite in sludge samples

For the first time, a procedure for the simultaneous determination of the iodinated drug amiodarone and its major metabolite, N-desethylamiodarone, in sludge from urban sewage treatment plants (STPs) is proposed. Matrix solid-phase dispersion (MSPD) followed by on-line cationic exchange clean-up, in modular configuration, was used as sample preparation technique. Liquid chromatography with tandem mass spectrometry (LC-MS/MS), based on a hybrid quadrupole time-of-flight (QTOF) system, was employed for the selective determination of target compounds. The optimized procedure provided exhaustive recoveries with little effect of the sample matrix in the efficiency of electrospray ionization (ESI). The overall recoveries of the method ranged between 95 and 1.11%, for samples spiked at different concentration levels. The achieved limits of quantification (LOQs) remained below 10 ng g(-1) for both compounds, and the linear response range extended up to 2500 ng g(-1). Amiodarone and N-desethylamiodarone were ubiquitous in sludge samples, from different STPs located in the Northwest of Spain, with maximum concentrations above 300 ng g(-1) referred to the freeze-dried matrix. They were also present in stabilized sludge (mixed with lime and thermally dehydrated), which is mostly disposed in agriculture fields as fertilizer. Furthermore, mono-iodinated analogues of amiodarone and N-desethylamiodarone were also tentatively identified in some samples from their accurate MS and MS/MS spectra. (C) 2015 Elsevier B.V. Summary: Organophosphate flame retardants (OPFRs) are increasingly used environmental pollutants due to restrictions on other flame retardants. Among them, the chlorinated OPFR tris(1,3-dichloro-2-propyl) phosphate (TDCPP) is of high environmental concern. Exposure of marine mussels to TDCPP for 28 days showed fast uptake and elimination, inhibition of AChE activity, and minimal proteomic effects in gills, with exposure time being the primary driver of protein abundance changes.

A new strategy for the computer-assisted development of reversed-phase liquid chromatography separation methods of unknown sample mixtures

Summary: A new strategy has been developed for computer-assisted methods in reversed-phase liquid chromatographic separations, using latent spectral information in chromatogram raw data files. By utilizing multivariate curve resolution-alternating least squares (MCR-ALS), this technique allows estimation of true spectra for individual components, providing a general approach for chromatography laboratory tools. This strategy is applicable to both known and unknown sample compositions, with the main difference being the need for an initial spectrum discovery process for unknown mixtures.

Functionalized mesoporous silicas SBA-15 for heterogeneous photocatalysis towards CECs removal from secondary urban wastewater

Summary: The photocatalytic activity of TiO2 nanoparticles supported on mesoporous silica SBA-15 was evaluated for the photodegradation of sulfadiazine. The 30% TiO2/SBA-15 sample showed the best performance in pure water solutions and also demonstrated higher SDZ degradation efficiency in urban secondary wastewater. Additionally, LC-MS analysis identified 65 other CECs in the wastewater sample, which were effectively removed by the photocatalytic system.

Summary: The primary focus of environmental toxicological studies is often on the direct effects of chemicals on exposed organisms, while the effects on subsequent non-exposed generations are often overlooked. In this study, the effects of simvastatin on a keystone crustacean were investigated, and it was found that simvastatin at environmentally relevant concentrations has significant inter and transgenerational effects on the neuroendocrine regulation of crustaceans, leading to adverse effects on reproduction and growth.

Screening of Contaminants of Emerging Concern in Surface Water and Wastewater Effluents, Assisted by the Persistency-Mobility-Toxicity Criteria

Summary: Contaminants of emerging concern (CECs) are a group of compounds with diverse origins that have recently garnered increasing environmental attention. The NOR-Water project aimed to identify the main CECs and their sources in the water environment of the Northern Portugal-Galicia transnational region. Through the use of liquid chromatography coupled to high resolution mass spectrometry (LC-HRMS), a total of 343 chemicals were tentatively identified in the analyzed samples, with 153 classified as persistent, mobile, and toxic (PMT) and 23 as very persistent and very mobile (vMvP).

Summary: This study investigated the generational effects of the antidiabetic drug MET on zebrafish and its nonexposed offspring. The results showed that MET had significant effects on reproductive-related parameters in zebrafish, with parental exposure to MET impacting critical molecular processes and upregulating male vtg1 expression, indicating an estrogenic effect. The nonexposed offspring also experienced negative effects on survival and growth due to parental MET exposure.

Summary: Pediatric Acute Myeloid Leukemia (AML) is a rare and heterogeneous disease characterized by gene fusions. Despite improved survival, a relapse still occurs in about 50% of patients. The NUP98-KDM5A chimeric protein is found exclusively in a specific subgroup of pediatric AML patients with poor prognosis. This study reveals that NUP98-KDM5A promotes genomic instability and contributes to malignant transformation.

Occurrence of persistent and mobile chemicals and other contaminants of emerging concern in Spanish and Portuguese wastewater treatment plants, transnational river basins and coastal water

Summary: This study examined the occurrence and fate of 52 emerging contaminants in transnational river basins, coastal areas, and wastewater treatment plants in Portugal and Spain. The results showed that these contaminants were widely present and were only partially removed by conventional treatment processes. These findings highlight the need for significant upgrades and coordination of wastewater treatment plants to meet future EU regulations.

Ozone membrane contactor to intensify gas/liquid mass transfer and contaminants of emerging concern oxidation

Summary: A tubular porous borosilicate membrane contactor was studied for ozone gas/water mass transfer and removal of contaminants in water. Increasing the gas and liquid flow rate improved the mass transfer efficiency, with the main resistances being in the water phase boundary layer and the membrane. The contactor showed good feasibility in removing contaminants, but photocatalytic ozonation did not significantly enhance treatment performance.

Breakthrough curve analysis of phosphorylated hazelnut shell waste in column operation for continuous harvesting of lithium from water

Multivariate surface self-assembly strategy to fabricate ionic covalent organic framework surface grafting monolithic sorbent for enrichment of aristolochic acids prior to high performance liquid chromatography analysis

Summary: In this study, an ionic covalent organic framework (iCOF) surface grafting monolithic sorbent was prepared for in-tube solid-phase microextraction (SPME) of aristolochic acids (AAs) in various samples. The density of quaternary ammonium ions in the iCOF was modulated for enhanced adsorption of AAs. The iCOF surface grafting monolithic sorbent was confirmed by different means. Several operating conditions were optimized to develop the online in-tube SPME-HPLC method, which showed good linearity, low limits of detection, and high recoveries for AAs in serum samples. This study presents a feasible approach for the application of COF-based monolithic sorbents in sample preparation.

Chitosan-alginate sponge with multiple cross-linking networks for adsorption of anionic dyes: Preparation, property evaluation, mechanism exploration, and application

Summary: A chitosan-alginate sponge with multiple cross-linking networks was developed for adsorption and enrichment of anionic dyes from food samples. The sponge showed better three-dimensional network structure, mechanical behavior, and stability compared to pure chitosan and alginate sponges. The adsorption performance and selectivity of the sponge were systematically studied, and it maintained high adsorption capacity and selectivity for anionic dyes. The sponge also showed satisfactory reusability and was successfully combined with high-performance liquid chromatography for the enrichment and detection of anionic dyes in candy and cocktail samples.

This study focused on Forsythia suspensa (Lian Qiao) and used metabolomics technology to explore its in vivo exposure. By comparing the differences before and after administration, a complete set of nontargeted analysis strategies for TCM exposure was established. The study identified 393 Lian Qiao-related chemical components in mice after administration, including prototypes and metabolites. This study provides a reference for research on the active ingredients of Lian Qiao in vivo.

A two-run heart-cut multidimensional gas chromatography method using flame ionization and mass spectrometry for automated and robust determination of nearly complete wine aroma-volatile profiles

Summary: A quantitative analytical method was developed and validated in this study to determine the concentrations of 81 aroma-relevant wine volatiles. The method utilizes stir bar sorptive extraction and multidimensional gas chromatography with flame ionization detector or mass spectrometer. The method demonstrated good linearity, reproducibility, and recovery, and was successfully applied to analyze 20 wine samples.
