Summary: Amarogentin (AMA) induces neurite outgrowth in PC12 cells through targeting the insulin receptor and activating downstream signaling pathways, including Ras/Raf/ERK and PI3K/AKT. Additionally, AMA is found to be involved in multiple neurotrophic pathways, with phosphorylation reactions potentially playing a key role in its neuritogenic activity.

Summary: In this study, a compound called inokosterone was isolated from Gentiana rigescens Franch, a Chinese herb medicine, and its antiaging effects were investigated using yeast and mammalian cells. Inokosterone extended the lifespan of yeast and enhanced the survival ability of mammalian cells. The mechanism of action involved antioxidative stress and autophagy. The results indicate that inokosterone exerts its antiaging activity by reducing oxidative stress and increasing autophagy.

Summary: In this study, a simple and rapid method for the correct identification of the geographical origins of Gentiana rigescens was developed. Marker components of iridoids were measured and used as a reference to characterize chemical profiles. The effects of climate factors on the content differences were examined. A novel two-dimensional correlation spectroscopy combined with deep learning was proposed for geographical origins identification. The results showed that this method provided a powerful tool for the cultivation and geographical origins identification of Gentiana rigescens.

Summary: GTS B, a dammaren-type triterpenoid glycoside from the traditional Chinese medicinal plant G. rigescens Franch, has shown significant anti-aging effects by inhibiting the TORC1/Sch9/Rim15/Msn signaling pathway and enhancing autophagy in yeast. This compound also increases the enzymatic activities of key anti-oxidative enzymes and improves survival rate under oxidative stress conditions.

Summary: This study investigates the different functions of ceramide in cellular differentiation and dedifferentiation. The results suggest that the function of ceramide heavily depends on its intracellular location, and further understanding of its function requires resolving its location during cellular status changes.

Summary: Biodegradable and lipid-like highly branched poly(beta-amino ester)s (HPAESA) were developed to enhance the biological functions of adipose-derived stem cells by gene transfection. HPAESA showed significantly higher efficiency and cell viability, as well as the ability to promote neurite outgrowth.

Summary: This study identified heat shock cognate 70 kDa protein (Hsc70) as the target protein of ABG-001 in PC12 cells, confirmed through inhibitors, cellular thermal shift assay (CETSA) and small-interfering RNA (siRNA) analysis. Inhibition of Hsc70 and siRNA significantly decreased ABG-001-induced neurite outgrowth, suggesting Hsc70 plays a key role in the neuritogenic activity of ABG-001.

Summary: Biochar, specifically rice husk biochar (RHB) and wood chip biochar (WCB), shows promise as a substitute for peat in horticultural plant production. In this study, the effects of RHB and WCB as components of a peat-based substrate on the growth and photosynthetic activity of Rhododendron delavayi seedlings were evaluated. Results showed that the addition of 20% WCB or 30-40% RHB enhanced the growth and photosynthetic activity of the seedlings, while the addition of 40% WCB had negative effects. The mixed substrates containing WCB or RHB had balanced porosity, suitable pH and electrical conductivity, and improved nutrient supply. This study suggests that a peat-based substrate with 20% WCB or 30-40% RHB is ideal for high-quality R. delavayi seedlings and sustainable horticulture production.

Summary: This study applied LIBS and CNN technologies to quickly identify the geographical origin of Gentiana rigescens Franch. Two methods were proposed to address the issue of too many variables in LIBS spectra, leading to improved discrimination accuracy. The results demonstrated the potential application of this combined method in quickly identifying the geographical origin of traditional Chinese medicine.

Summary: This study aimed to analyze the effect and mechanism of Wnt5a-loaded fibrin hydrogel on a 10-mm rat sciatic nerve defect. Results showed that Wnt5a could promote Schwann cell proliferation and the expression of various neurotrophic factors, and the Wnt5a/Fn group outperformed the control groups in terms of nerve regeneration and functional recovery 12 weeks after the operation. The Wnt5a/Fn nerve conduit may promote peripheral nerve defect regeneration by facilitating multiple neurotrophin secretion, combining vascularization and neurotrophic growth cues.
