This paper reports the synthesis and characterization of a new class of tetrabenzoporphyrins bearing glucosyl or polyamine units on meso positions to improve the targeting of cancer cells. Photocytotoxic activity of these photosensitizers was tested on cell lines HaCaT and MCF-7 and compared to Photofrin II (R). (C) 2009 Elsevier Ltd. All rights reserved.

Synthesis and Biological Evaluation of Benzo[b]thiophene Acylhydrazones as Antimicrobial Agents against Multidrug-Resistant Staphylococcus aureus

Summary: In this study, new series of compounds were synthesized by combining the benzo[b]thiophene nucleus and the acylhydrazone functional group for screening against Staphylococcus aureus. Among the compounds tested, (E)-6-chloro-N'-(pyridin-2-ylmethylene)benzo[b]thiophene-2-carbohydrazide (II.b) showed promising antimicrobial activity with a minimal inhibitory concentration of 4 μg/mL against S. aureus strains, including clinically isolated strains resistant to methicillin and daptomycin.

Antibacterial Photodynamic Therapy in the Near-Infrared Region with a Targeting Antimicrobial Peptide Connected to a p-Extended Porphyrin

Summary: The increase of antimicrobial resistance is a major health problem worldwide, and new bactericidal strategies are needed. Antimicrobial photodynamic therapy (a-PDT) can generate reactive oxygen species to target cellular structures, reducing the risk of bacterial resistance. A new conjugate consisting of an antimicrobial peptide and a pi-extended porphyrin photosensitizer has been designed to effectively kill both Gram-positive and Gram-negative bacteria when excited by near IR. Moreover, this conjugate can achieve a low minimum bactericidal concentration without causing toxicity to skin cells.

Ammonium Acetate Catalyzed Formation of 1,5-Benzodiazepines through [4+2+1] Cycloaddition Involving 5-Hydroxymethylfurfural

Summary: The use of the renewable platform molecule 5-HMF in multi-component reaction with a diamine and an alkynone to generate seven-membered 1,5-benzodiazepines is described. Ammonium acetate in ethanol proved to be a clean and efficient system among catalysts/solvents couples. The optimized conditions were used to investigate the scope of the reaction providing a library of 1,5-benzodiazepines in moderate to good yields from a wide range of diversely substituted diamines and alkynones or alkyl alkynoates.

Importance of the 2,6-Difluorobenzamide Motif for FtsZ Allosteric Inhibition: Insights from Conformational Analysis, Molecular Docking and Structural Modifications

Summary: A conformational analysis and molecular docking study compared 2,6-difluoro-3-methoxybenzamide (DFMBA) with 3-methoxybenzamide (3-MBA) to investigate the effect of fluorination on FtsZ inhibition and anti S. aureus activity. The presence of fluorine atoms in DFMBA resulted in a non-planar conformation, making it more adaptable to the reported co-crystallized complexes with FtsZ. Molecular docking studies showed strong hydrophobic interactions between the difluoroaromatic ring and residues Val203, Val297, and Asn263, as well as critical hydrogen bonds with residues Val207, Leu209, and Asn263. Changing the carboxamide functional group to benzohydroxamic acid or benzohydrazide rendered the compounds inactive, emphasizing the importance of the carboxamide group.

Shining a Light on Prostate Cancer: Photodynamic Therapy and Combination Approaches

This review provides an overview of conventional treatments, principles of PDT, different types of photosensitizers, and ongoing research on combination therapy for prostate cancer. PDT has the potential to be a more effective and less invasive treatment option, and research aims to improve its selectivity and efficacy.

In Silico Identification of Potential Inhibitors of the SARS-CoV-2 Main Protease among a PubChem Database of Avian Infectious Bronchitis Virus 3CLPro Inhibitors

Summary: Comparative analysis of amino acid sequences and 3D structural alignment revealed significant structural homologies between the main proteases of SARS-CoV-2 and IBV. In silico screening using a PubChem BioAssay database identified four covalent inhibitors and five non-covalent inhibitors for SARS-CoV-2 protease. Molecular dynamics simulations further investigated the binding of these inhibitors, and predictive ADMET calculations suggested promising pharmacokinetic properties for the nine compounds.

Xylan-Porphyrin Hydrogels as Light-Triggered Gram-Positive Antibacterial Agents

Summary: This study presents a synthesis method for light-triggered antibacterial hydrogels based on xylan chains chemically bound to meso-tetra(4-carboxyphenyl)porphyrin (TCPP). TCPP not only acts as an efficient photosensitizer against Gram-positive bacteria, but also serves as a cross-linking agent for the hydrogels. The hydrogels were characterized using infrared spectroscopy (ATR-FTIR), scanning electron microscopy (SEM), and swelling and rheological tests. Under visible light irradiation, the hydrogels showed interesting antimicrobial activity against Staphylococcus aureus and Bacillus cereus, suggesting their potential for the treatment of skin bacterial infections through photodynamic antimicrobial chemotherapy (PACT).

Summary: Glycosteroids, including synthetic monocatenar carbohydrate steroid hybrids, have been synthesized by connecting carboxymethylglycosyl lactone-based allyl amides with allylated steroids through cross-metathesis reaction. The reaction successfully utilized glucose, galactose, cellobiose, and dihydrocholesterol as substrates, resulting in the synthesis of variously protected new glycosteroids.
