Susceptibility of Trypanosoma evansi to cordycepin

Drugs, which are effective during the early stage of trypanosomosis, but poorly penetrate the blood-brain barrier, are ineffective when parasites reach the brain and cause encephalitis. In order to seek alternative treatments, the aim of this study was to test the susceptibility of T. evansi to cordycepin in vitro and in rats experimentally infected. In vitro, a significant decrease (P < 0.01) in live trypanosomes in the concentrations of 5.0 and 10 mu g/mL was observed 1 hour after the beginning of the study, as well as at 3, 6, 9 and 12 hours in all concentrations compared to control. Although no curative effects were observed in the in vivo assay in the majority of groups, the drug was able to maintain parasitemia at low levels, therefore increasing the longevity of rats when compared to positive control group. Rats that received cordycepin alone or in combination with adenosine deaminase inhibitor (ADA: EHNA hydrochloride), did not show trypomastigote forms of the parasite in the bloodstream 24 hours after the administration. These animals remained negative in blood smears on average for 8 days, but thereafter had a recurrence of parasitemia. Among all the infected animals, only three rats in the group treated with the combination of cordycepin (2 mg/kg) and EHNA hydrochloride (2 mg/kg) remained negative during the experimental period. The curative efficacy of 42.5% was confirmed by PCR using T. evansi-specific primers. Thus, we conclude that cordycepin has biological effect against T. evansi, as previously reported in infections by T. brucei, T. cruzi and Leishmania sp. The treatment with cordycepin, when protected by an inhibitor of ADA, can prolong the survival of T. evansi-infected rats and provide curative efficacy. (C) 2011 Elsevier Masson SAS. Global prevalence of free-living amoebae in solid matrices - A systematic review with meta-analysis

Summary: The ubiquitous free-living amoebae (FLA) are microorganisms of significant medical, sanitary, and ecological importance. However, their characterization within solid matrices such as soil, dust, sediment, mud, sludge, and compost remain to be systematized. In this study, a systematic review with meta-analysis was conducted to explore the global distribution of FLA in solid matrices. The analysis of 104 out of 4,414 scientific articles revealed that FLA prevalence in solid matrices was 55.13% globally. Soil had the highest prevalence (72.40%), followed by sediment (57.91%), mud (52.90%), dust (48.60%), and sewage sludge (40.19%). Acanthamoeba spp. (52.23%) and Hartmanella/Vermamoeba spp. (36.06%) were the most prevalent FLA species.

Drosophila melanogaster as a model of systemic dermatophytosis

Summary: This study validates Drosophila melanogaster flies as a suitable model to study dermatophytic infections. It shows that wild-type flies are resistant to infection, while Toll-deficient flies are susceptible. Antifungal drugs can protect flies from infection, except for one specific fungus.

Detection of paratuberculosis in dairy cows from southern Brazil

Indirect ELISA tests and PCR were performed on milk and fecal samples, respectively. The results indicated that paratuberculosis is likely endemic in the northwest and northeast mesoregions.

Blood concentrations of calcium, phosphorus and magnesium and dietary cation-anion difference in dairy cows

Summary: The study aimed to investigate the impact of pre and postpartum DCAD on serum calcium, phosphorus, and magnesium levels in peripartum Holstein cows. Blood samples were collected from 56 cows (18 primiparous and 38 multiparous) on different days relative to parturition to analyze mineral levels, while diet and urine samples were also collected. Prepartum DCAD showed a positive correlation with total and corrected calcium levels at day-7, and a negative correlation with magnesium levels at day-7. However, prepartum DCAD did not significantly affect phosphorus levels. Postpartum DCAD did not have any correlation with serum mineral levels. The study highlights the importance of considering parity and time period when evaluating mineral levels in Holstein cows during the transition period.

Clioquinol and 8-hydroxyquinoline-5-sulfonamide derivatives damage the cell wall of Pythium insidiosum

Summary: This study evaluates the antimicrobial activity of three 8-hydroxyquinoline derivatives against Pythium insidiosum and determines their pharmacodynamic characteristics. Results show that Clioquinol, PH151, and PH153 are active against all isolates, with MIC values ranging from 0.25 to 2 μg/mL. They also exhibit a time- and dose-dependent antimicrobial effect, damaging the P. insidiosum cell wall. These findings reinforce the potential of 8-hydroxyquinoline derivatives as new drugs for treating pythiosis.
