hTERT methylation and expression in gastric cancer

Gastric cancer is the second most prevalent cause of cancer death worldwide. DNA methylation is a common event in gastric carcinogenesis. hTERT seems to be the rate-limiting determinant of telomerase activation, which is responsible for stability and life span. hTERT hypermethylation has been associated with telomerase expression. In the present study, we investigated the promoter methylation status and hTERT protein expression in gastric cancer and normal mucosa samples. One hundred and nine gastric cancer and 53 normal mucosa samples were investigated through methylation-specific PCR. Immunohistochemistry was analysed using peroxidase in 55 gastric cancer and 18 normal gastric mucosa samples. This is the first study evaluating hTERT methylation status in gastric carcinogenesis. We did not observe hTERT protein expression in normal gastric mucosa. Moreover, hTERT expression was observed in 80% of tumours and was associated with gastric cancer (p < 0.0001). Partial methylation was the most frequent pattern in gastric samples, even in normal mucosa. The frequency of specimens presenting hypermethylation was significantly higher in tumours than in normal mucosa samples (p = 0.0002), although the presence of hypermethylated promoter was not associated with a higher frequency of hTERT expression. A low correlation between hTERT protein expression and methylation was verified in gastric cancer samples. There was a clear difference in the frequency of hTERT expression and methylation within tumoral and non-tumoral tissues. Methylation status and telomerase expression may be useful for the diagnosis of gastric cancer and may have an impact on the anti-telomerase strategy for cancer therapy.

Understanding these variants can contribute to the diagnosis and pathophysiology of autism in Amerindians and admixed populations.

ZDHHC9 X-linked intellectual disability: Clinical and molecular characterization

Summary: The ZDHHC9 gene mutation leads to intellectual disabilities and symptoms such as low IQ, developmental delay, facial dysmorphisms, and skeletal defects. Although there are differences in clinical manifestations among different patients, intellectual disabilities and developmental delay are reported in all cases. These mutations mainly occur in the arginine residues located in the cytoplasmic domains of the protein and may be hotspot mutations.

Summary: Gliomas are common malignant brain tumors with limited treatment options. Plant-derived quinone derivatives, such as eleutherine, have shown potential anti-glioma activity. This study found that eleutherine inhibits glioma cell proliferation, migration, and invasion, and induces apoptosis. These results suggest the potential clinical use of eleutherine for treating gliomas.

Histone Modifications in Alzheimer's Disease

Summary: Due to the combination of genetic variants and environmental factors, epigenetic modifications are predicted to play a role in the etiopathology of Late-onset Alzheimer's disease (LOAD). This review highlights the main histone modifications and their functional role, including histone acetylation, methylation, and phosphorylation, and discusses changes in these modifications during the aging process and in Alzheimer's disease (AD). The review also discusses the potential use of epigenetic drugs, such as histone deacetylase (HDAC) inhibitors, for the treatment of AD.

Summary: Alzheimer's disease is a progressive neuroinflammatory and neurodegenerative disorder that affects different regions of the brain. The accumulation of beta-amyloid protein, formation of neurofibrillary tangles, and inflammatory processes contribute to its pathophysiology. CDK10 and CDK11 have high expression in AD patients compared to control, and they present a positive correlation of gene expression in the analyzed groups and tissues, suggesting their important role in the pathogenesis of AD.

Study of Genotoxicity, Activities on Caspase 8 and on the Stabilization of the Topoisomerase Complex of Isoeleutherin and Analogues

Summary: This study evaluated the genotoxicity of Ethanol Extract (EEEp), Dichloromethane Fraction (FDCMEp) and isoeleutherin isolated from Eleutherine plicata, using the micronucleus test and molecular docking. Isoeleutherin and analogues were subjected to in silico toxicity prediction, and compounds free of toxicological risks (CP13, CP14, CP17 and isoeleutherin) were selected for molecular docking in Topoisomerase II. The results showed that the structural changes of isoeleutherin led to the obtaining of a molecule with a lower mutagenic potential, and the CP13 can be considered a prototype compound for the development of new molecules with pharmacological potential.

Association between Polymorphism of Genes IL-1A, NFKB1, PAR1, TP53, and UCP2 and Susceptibility to Non-Small Cell Lung Cancer in the Brazilian Amazon

Summary: A study conducted in the Brazilian Amazon found that five genetic variants (IL-1A, NFKB1, PAR1, TP53, and UCP2) were significantly associated with the development of non-small cell lung cancer (NSCLC). Variants in NFKB1, PAR1, and TP53 were negatively correlated with NSCLC, while variants in IL-1A and UCP2 were positively correlated. These findings suggest the importance of genetic variations in NSCLC susceptibility.

The findings suggest that the GG genotype of the rs10739971 variant is associated with a lower risk of gastric cancer. This is the first study to report this association in this unique population.

Summary: Migrant birds prepare differently for spring breeding and autumn migration. By analyzing gene expressions in different tissues, this study found significant changes in glial cells and neurogenesis in the brain of semipalmated sandpipers during the wintering period. The gene expressions in newly arrived and premigratory birds were associated with neurogenesis, metabolic pathways, glial changes, immune response, inflammatory cytokines, and cell death pathways.
