Lung Perfusion Imaging with Monosized Biodegradable Microspheres

After intravenous injection, particles larger than red blood cells will be trapped in the first capillary bed that they encounter. This is the principle of lung perfusion imaging ill nuclear medicine, where macroaggregated albumin (MAA) is radiolabeled with (99m)Tc, infused into a patient's arm vein, and then imaged with gamma scintigraphy. Our aim was to evaluate if monosized microspheres Could replace (99m)Tc-MAA. Biodegradable poly(L-lactide) microspheres containing chelating bis(picolylamine) end groups were prepared by a flow focusing method on a microfluidic glass chip and were of highly homogeneous size (9.0 +/- 0.4 mu m), The microspheres were radiolabeled with [(99m)Tc(H(2)O)(3)(CO)(3)](+) and then evaluated in mice for lung perfusion imaging. Fifteen minutes after injection, 79.6 +/- 3.8% of the injected activity was trapped in the lungs of mice. Monosized biodegradable radioactive microspheres are, thus, appropriate lung perfusion imaging agents. Other sizes of these highly uniform microspheres have the potential to improve diagnostic and therapeutic approaches in diverse areas of medicine.

Summary: This study found that LPNs showed a prolonged and enhanced gene silencing effect compared to LPXs after pulmonary administration, possibly due to the sustained release characteristics of PLGA and the more stable retention of LPNs in lung tissue. Additionally, the presence of PLGA effectively alleviated the acute inflammatory effect of cationic lipids on the lungs. The findings suggest that PLGA-based LPNs may be an effective formulation strategy for sustained gene silencing effects in the lung via pulmonary administration.

Summary: Albumin microparticles, particularly macroaggregated albumin (MAA), are commonly used in pharmaceutical applications for drug delivery and imaging. The microfluidic chip offers an elegant and controllable method for preparing albumin microparticles for biomedical applications.

SPECT/CT Imaging of 111Ag for the Preclinical Evaluation of Silver-Based Antimicrobial Nanomedicines

Summary: Single-photon emission computed tomography (SPECT) is a useful tool for noninvasively visualizing the biodistribution of silver-containing compounds in rodents. The study found that antimicrobial silver nanoparticles, formed by neutron irradiation of palladium foil, can be stably incorporated into the lungs of mice without significant uptake in other organs. In a mouse model of pulmonary Pseudomonas aeruginosa infection, the nanoparticles effectively reduced bacterial burden without inducing excessive inflammation.

Potential of Nuclear Imaging Techniques to Study the Oral Delivery of Peptides

Summary: Peptides are small biomolecules that can stimulate or inhibit important functions in the human body. However, the clinical use of peptides through oral delivery is limited due to their sensitive structure and physiological barriers in the gastrointestinal tract. These barriers can be overcome by chemical and mechanical approaches, and nuclear imaging provides a better platform for observing the gastrointestinal transit and quantitative distribution of radiolabeled peptides. This review provides a brief background on the oral delivery of peptides and discusses how nuclear imaging can aid in the study of their gastrointestinal transit.

Preclinical PET Imaging and Toxicity Study of a 68Ga-Functionalized Polymeric Cardiac Blood Pool Agent

Quantitative Evaluation of a Multimodal Aptamer-Targeted Long-Circulating Polymer for Tumor Targeting

Summary: This study presents a modular conjugation strategy based on click chemistry for the modification of high molecular weight hyperbranched polyglycerol (HPG) with sgc8 aptamer, fluorescent dyes, and In-111. The results indicate strong affinity of sgc8 against solid tumor-derived cell lines, but nonspecific uptake of scrambled ssDNA-functionalized HPG in cells highlights inherent challenges of aptamer-targeted probes. The study validates HPG-sgc8 as a nontoxic nanoprobe with high affinity against breast and lung cancer cells, and shows increased stability compared to free sgc8.
