Summary: This study conducted a comprehensive systematic review and meta-analysis to investigate the efficacy and safety of autologous hematopoietic stem-cell transplantation (AHSCT) in patients with multiple sclerosis (MS). The results showed that AHSCT led to a significant decrease in Expanded Disability Status Scale (EDSS) score and annualized relapse rate. Most patients remained relapse-free after AHSCT and the overall survival rate was high.

Summary: The effect of autologous hematopoietic stem cell transplantation (auto-HSCT) versus conventional chemotherapy or allogeneic hematopoietic stem cell transplantation (allo-HSCT) on the survival of patients with advanced follicular lymphoma (FL) is uncertain. This study included 13 studies and found that auto-HSCT improved overall survival (OS), progression-free survival (PFS), and event-free survival (EFS) compared with conventional chemotherapy. In addition, auto-HSCT resulted in longer OS and lower non-relapse mortality (NRM) compared with allo-HSCT.

Summary: A systematic review and meta-analysis evaluated the outcomes of mismatched unrelated donor hematopoietic stem cell transplantation (MMUD-HSCT). The results showed that MMUD-HSCT had favorable treatment outcomes and an acceptable toxicity profile in patients lacking HLA-matched or haploidentical donors, thus expanding the accessibility of HSCT to underrepresented populations.

Summary: NK cells play a vital role in immune reconstitution after allogeneic hematopoietic stem cell transplantation. Higher NK cell reconstitution and graft NK cell content are associated with better overall survival, lower relapse rate, and lower acute GVHD incidence. Pre-emptive NK cell infusions may prevent disease relapse post-HSCT.

Summary: This article presents a systematic review and meta-analysis that suggests hematopoietic stem cell transplantation (HSCT) can be an effective treatment for refractory chronic inflammatory demyelinating polyneuropathy (CIDP). The study found a high responsiveness rate and significant reduction in the use of immune modulating or suppressive drugs after HSCT.

Summary: Multiple sclerosis (MS) is a chronic disease of the central nervous system that can lead to lifelong disability. Despite various treatment options, some patients still face severe symptoms and long-term disability risk. Recent studies have shown that autologous hematopoietic stem cell transplantation (AHSCT) is a highly effective and relatively safe treatment option for highly active MS.

Summary: Cardiovascular diseases are a significant cause of mortality and morbidity among survivors of hematopoietic stem cell transplantation (HSCT), but there is a lack of detailed description on the incidence of cardiovascular events (CVEs) in this population. This systematic review provides a summary of the evidence on the incidence of CVEs in HSCT recipients. The review includes 57 studies and found that the incidence of CVEs after autologous HSCT was higher compared to allogeneic HSCT, with arrhythmia being a contributing factor. Long-term CVEs were also observed in both types of HSCT survivors. CVEs remain an important but under-reported cause of morbidity and mortality in HSCT recipients.

Summary: Front-line high-dose therapy followed by autologous stem cell transplantation (ASCT) is still a beneficial approach for newly diagnosed multiple myeloma (MM) patients in the era of novel agents. However, the benefit of HDT/ASCT in terms of progression-free survival (PFS) and overall survival (OS) remains inconsistent. We conducted a systematic review and meta-analysis to evaluate the advantages of upfront HDT/ASCT and identified factors influencing survival outcomes.

Summary: Hematopoietic stem cell transplant recipients are susceptible to Clostridium difficile infection. A meta-analysis of relevant studies found an overall CDI prevalence of 13.2% in HSCT patients, with higher rates among allogeneic transplantation recipients compared to autologous HSCT recipients. The incidence rates of CDI varied worldwide, with North America having higher rates than Europe but no significant difference with Asia. PCR-diagnosed CDI had a significantly higher prevalence than enzyme immunoassay-diagnosed CDI. CDI was a common complication among HSCT recipients, with recurrence and severe cases observed.

Summary: The optimal consolidation strategy for newly diagnosed primary central nervous system lymphoma (PCNSL) remains controversial between autologous hematopoietic cell transplant (auto-HCT) and whole-brain radiotherapy (WBRT). A systematic review and meta-analysis of randomized controlled trials was conducted, revealing no significant differences in survival outcomes between auto-HCT and WBRT consolidation, but a higher degree of neurocognitive decline associated with WBRT. Individualized decision-making based on age, frailty, and co-morbidities is recommended.

Summary: The co-infusion of mesenchymal stem cells (MSCs) in allogeneic hematopoietic stem cell transplantation (allo-HSCT) has shown improvement in engraftment and reduced risk of chronic graft-versus-host disease (cGVHD). However, the effects varied among different age and disease groups, suggesting potential benefits for certain populations.

Summary: While the incidence of acute kidney injury (AKI) in children after hematopoietic stem cell transplantation (HCT) has not been fully addressed, a systematic literature review revealed an estimated incidence of approximately 51%. The incidence of AKI varied depending on the criteria used for evaluation, ranging from 50% to 60%. However, there was no significant correlation between the years of publication and the incidence of AKI. In conclusion, AKI affects approximately half of the pediatric population undergoing HCT, and it is expected to decrease with advancements in medical techniques.

Summary: This study systematically evaluated and compared the efficacy and safety of consolidative HSCT after CD19 CAR-T therapy with non-HSCT in the treatment of ALL. The results showed that consolidative HSCT after CD19 CAR-T therapy could prolong OS and LFS and reduce the risk of relapse, with acceptable incidence rates for adverse events. More high-quality randomized controlled trials are needed to further determine the efficacy of HSCT.

Summary: Autologous hematopoietic cell transplantation has been the standard approach for patients with relapsed/refractory diffuse large B cell lymphoma. However, the introduction of CAR T cell therapy has changed the management of these patients, especially with the approval of CD19-directed CAR-T therapy in the second-line setting for high-risk groups. The American Society of Transplantation and Cellular Therapy has formulated consensus recommendations to address the lack of consensus on the role and sequencing of HCT and cellular therapies in DLBCL.
