This experiment was conducted to examine the effect of dietary copper supplementation on somatostatin (SS) and growth hormone-releasing hormone (GHRH) mRNA expression levels in the hypothalami of growing pigs. A total of 45 crossbred pigs were randomly assigned to three groups of 15 pigs each; five replicates of three animals comprised each group. Pigs were allocated to diets that contained 5 mg/kg copper (control), 125 mg/kg copper sulfate, or 125 mg/kg copper methionine. At the end of the experiment, five pigs were selected at random from each group and slaughtered, and hypothalami were collected for determination of SS and GHRH mRNA expression levels. The results showed that the SS expression levels were lower and the GHRH levels were higher in pigs fed the diets with 125 mg/kg copper methionine (P < 0.05) and 125 mg/kg copper sulfate (P < 0.05), respectively, than in pigs fed the diet with 5 mg/kg copper. Furthermore, the relationship between SS mRNA and GHRH mRNA abundance had a significantly negative correlation (P < 0.05). The data indicated that high dietary copper could enhance GHRH mRNA expression levels and suppress SS mRNA expression levels in the hypothalami of pigs. High lever dietary copper (125 mg/kg copper sulfate or 125 mg/kg copper methionine) increased pigs' growth performance and feed efficiency but had no significant effect on daily feed intake; 125 mg/kg copper sulfate or 125 mg/kg copper methionine at the same lever had no difference on growth promoting in pigs.

Sirtuin 3 inhibits nuclear factor-KB signaling activated by a fatty acid challenge in bovine mammary epithelial cells

Lei Liu, Hong Lu, Juan J. Loor, Ahmad Aboragah, Xiliang Du, Jianhua He, Tao Peng, Jianming Su, Zhe Wang, Guowen Liu, Xinwei Li

Summary: Susceptibility to mastitis is highest during the peri-partal period, with factors like ketosis exacerbating the condition. The study found that ketotic cows had lower levels of SIRT3 and higher activation of NF-Kappa B signaling in the mammary gland. This suggests that SIRT3 could be a promising target for alleviating localized inflammation in the mammary gland caused by exposure to high levels of NEFA.

Sirtuin 3 relieves inflammatory responses elicited by lipopolysaccharide via the PGC1α-NFκB pathway in bovine mammary epithelial cells

Lei Liu, Baogen Wang, Wei Yang, Qianming Jiang, Juan J. Loor, Lu Ouyang, Huilun Tang, Renxu Chang, Tao Peng, Chuang Xu

Summary: Excessive inflammation in bovine mammary endothelial cells (BMEC) due to mastitis leads to disease progression and eventual culling of cattle. Sirtuin 3 (SIRT3), a mitochondrial deacetylase, downregulates pro-inflammatory cytokines in BMEC exposed to high concentrations of nonesterified fatty acids by blunting nuclear factor-KB (NFKB) signaling. The study investigated the effects of SIRT3 and PGC1 alpha on inflammatory responses in bovine mammary epithelial cells and confirmed the potential benefits of targeting SIRT3 to prevent or alleviate negative effects of mastitis.

Strontium Attenuates Hippocampal Damage via Suppressing Neuroinflammation in High-Fat Diet-Induced NAFLD Mice

Shuai Wang, Fangyuan Zeng, Yue Ma, Jiaojiao Yu, Chenyao Xiang, Xiao Feng, Songlin Wang, Jianguo Wang, Shanting Zhao, Xiaoyan Zhu

Summary: The study found that strontium (Sr) has a protective effect on hippocampal damage in non-alcoholic fatty liver disease (NAFLD) mice. Sr can reduce the expression of caspase-3 by suppressing endoplasmic reticulum stress (ERS), and inhibit neuroinflammation and the expression of inflammatory cytokines, thereby alleviating NAFLD-induced hippocampal damage. This finding suggests that Sr could be a potential candidate for protecting against neural damage caused by NAFLD.

Untargeted metabolomics and lipidomics to assess plasma metabolite changes in dairy goats with subclinical hyperketonemia

Yan Huang, Yezi Kong, Bingyu Shen, Bowen Li, Juan J. Loor, Panpan Tan, Bo Wei, Linshan Mei, Zixin Zhang, Chenxu Zhao, Xiaoyan Zhu, Simeng Qi, Jianguo Wang

Summary: This study comprehensively assessed the metabolomic profiles of dairy goats with subclinical hyperketonemia (SCHK) and found that SCHK goats had higher concentrations of nonesterified fatty acids, beta-hydroxybutyrate, and aminotransferase, but lower glucose concentrations compared to clinically healthy goats. Pathway enrichment analysis revealed abnormalities in the citrate cycle, alanine, aspartate and glutamate metabolism, and phenylalanine metabolism in SCHK goats. Overall, the altered metabolites in SCHK goats indicated a more severe negative energy balance and an imbalance in the tricarboxylic acid cycle, lipid metabolism, and amino acid metabolism. These findings provide a more comprehensive understanding of the pathogenesis of SCHK in dairy goats.

High-fat diet-disturbed gut microbiota-colonocyte interactions contribute to dysregulating peripheral tryptophan-kynurenine metabolism

Penghao Sun, Mengli Wang, Yong-Xin Liu, Luqi Li, Xuejun Chai, Wei Zheng, Shulin Chen, Xiaoyan Zhu, Shanting Zhao

Summary: This study found that a high-fat diet disrupts the interactions between gut microbiota and colonocytes, leading to dysfunction in the Trp-Kyn metabolism pathway. The high-fat diet promotes the expansion of Proteobacteria and activates the IDO1-mediated Kyn pathway, causing Trp depletion and Kyn accumulation in the circulation. However, supplementation with butyrate restores mitochondrial function and remodels the gut microbiota, improving serum Kyn accumulation.
