Summary: The study demonstrated that Lp31-modified Caelyx exhibited enhanced cytotoxicity and cellular uptake in C26 cell line compared to Caelyx. In BALB/c mice bearing C-26 murine carcinoma, Lp31-modified Caelyx groups showed significantly higher doxorubicin concentration at tumor tissue. Furthermore, Lp31-modified Caelyx at the dose of 10 mg/kg resulted in significant tumor growth inhibition and enhanced survival time compared to Caelyx.

Summary: The study developed new liposomal formulations to enhance the anti-cancer therapeutic application of Galbanic acid, and demonstrated that RGD peptide decoration could improve targeted delivery to tumor vasculature. The results showed that the targeted liposomes could enhance receptor-mediated endocytosis both in vitro and in vivo, leading to increased accumulation in colon tumors.

Summary: The study successfully prepared EXO-DTX with a particle size of 149.5 nm, showing significantly greater cytotoxicity than DTX monomers and better targeting and cellular uptake both in vitro and in vivo. The potential of tumor cell-derived exosomes as a drug delivery system targeting the parent cancer was also explored, suggesting exosomes might be used as a potential antitumor drug delivery system (DDS).

Summary: The FDA granted accelerated approval to selpercatinib on May 8, 2020 for patients with metastatic RET fusion-positive non-small cell lung cancer, RET-mutant medullary thyroid cancer, and RET fusion-positive thyroid cancer. The approval was based on significant effects on overall response rate and prolonged duration of responses observed in clinical trials across multiple indications. The product label includes warnings for various adverse reactions such as hepatotoxicity, hypertension, and embryo-fetal toxicity.

Summary: Active targeting of the tumor microenvironment using nucleolin-targeted PEGylated liposomal doxorubicin (AS-PLD) has shown higher therapeutic efficacy compared to traditional liposomal doxorubicin (PLD). In vitro studies demonstrated that AS-PLD exhibited increased cytotoxicity and cellular uptake in cancer cells. Additionally, competition assays confirmed the specificity of AS-PLD in targeting tumor cells. Furthermore, in vivo analysis showed higher accumulation of AS-PLD in the tumor after 72 hours post-injection, indicating its potential as an active-targeted formulation for cancer treatment.

Summary: In this study, diselenide-crosslinked nanoparticles were successfully fabricated for doxorubicin delivery. The nanoparticles exhibited desirable morphology and size distribution, and the starting material used improved biocompatibility and physiological stability. The high drug encapsulation efficiency of the nanoparticles was achieved through electrostatic interactions between doxorubicin and the material. In vitro and in vivo experiments demonstrated that the doxorubicin-loaded diselenide-crosslinked nanoparticles had higher drug accumulation and enhanced growth inhibition compared to the control samples.

Summary: In this study, a liver-targeting ligand was conjugated with polyethylene glycol and incorporated into liposomes to improve the delivery of the chemotherapy drug for hepatocellular carcinoma (HCC). The optimized formulation showed high drug encapsulation capacity and demonstrated superior toxicity against HCC cells compared to the commercial formulation, while being less toxic to non-cancerous cells. It also exhibited enhanced tumor penetration and antitumor efficacy in a mouse liver cancer model.

Summary: Two liposomal formulations, one of which is coated with hyaluronic acid, have been developed to enhance the solubility of Thymoquinone (TQ) and increase its absorption at the cellular level, especially in the nucleus. These formulations not only decrease TQ toxicity observed at high doses in cells, but also result in a gradual and prolonged release of TQ in both coated and uncoated liposomes.
