Summary: The study found diverse drug-resistant strains of Mycobacterium tuberculosis in Oman, with drug resistance-conferring mutations widespread in multiple spoligo-lineages, indicative of a large resistance reservoir. The overall resistance rates among Omanis and expatriates were similar, but multi-drug resistance and Pyrazinamide resistance were significantly higher among Omanis, while Isoniazid resistance was greater among expatriates.

The rpoB gene had the highest number of mutations, and the Beijing spoligotype was the most prevalent. Summary: This study aimed to investigate the population structure of Mycobacterium tuberculosis (MTB) genotypes in Fujian and explore the risk factors associated with Beijing genotype infection. The study also examined the relationship between Beijing genotype and drug resistance. The results showed that the Beijing genotype is the predominant sublineage in Fujian and its prevalence decreases with advancing patient age. Furthermore, the emergence of novel spoligotypes of highly drug-resistant MTB isolates highlights the need for ongoing molecular genotyping surveillance in the province.

Summary: The aim of this study was to investigate the genetic variability of the insertion sequence (IS) 6110 in the Mycobacterium tuberculosis complex (MTBC). The results showed a high mutation rate in IS6110, with low-copy-number strains having higher mutation rates than high-copy-number strains. Additionally, specific IS6110 variants were found in certain strains, and Haarlem and X family strains shared a common IS6110 copy in a specific location.

However, further large-scale research is needed to confirm these findings.

Summary: Recent molecular epidemiological data indicate significant genetic variation within different lineages of the Mycobacterium tuberculosis complex (MTBC), with specific adaptations to different human populations. This study in the Volta Region of Ghana revealed a diverse population structure of clinical MTBC isolates, dominated by M. tuberculosis sensu stricto (MTBss) and M. africanum (MAF) lineages. The presence of various spoligotype patterns and distinct Lineage 4 sub-lineages within the MTBC isolates further highlights the genetic diversity and localization of specific lineages in this region.

Summary: The whole-genome sequence comparison of the MTC genomic sequences identified a unique gene, CRISPR-csm4, which can be used directly on clinical samples to detect MTC and differentiate between MTC species, lineages, and sublineages. The PCR-based detection of MTC in clinical samples was confirmed by testing the CRISPR-csm4 gene on a collection of MTC and non-tuberculous mycobacteria isolates. The results showed that CRISPR-csm4 can accurately detect MTC isolates.
