Bacillus thuringiensis (Bt) Berliner is a promising agent for microbial control of agriculturally and medically important insects. This study aimed at searching for Bt strains encoding Cry proteins that act more efficiently against fall armyworm. Thirty Bt strains were isolated from soil samples in Pernambuco State and evaluated through bioassays. Among these, strain I4A7 was the most efficient against the fall armyworm, Spodoptera frugiperda (J. E. Smith, 1797) (Lepidoptera: Noctuidae), and thus it was characterized by biochemical sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and molecular (polymerase chain reaction (PCR) and sequencing reaction) methods. The protein pattern of this strain on a SDS-PAGE was similar to that of B. thuringiensis israelensis (Bti). Moreover, I4A7 cry DNA sequence showed high identity (99-100%) to genes cry4Aa, 4Ba, 10Aa, 11Aa, cyt1Aa and cyt2B from Bti. The toxicity of the newly isolated Bti-like strain upon S. frugiperda should be considered as this strain might be used in combination with other Bt strains, such as B. thuringiensis var. kurstaki (Btk).

Marianna de Carvalho Climaco, Yrna Lorena Matos de Oliveira, Anne Caroline Santos Ramos, Jucicleide Ramos-de-Souza, Audrey Rouse Soares Tavares Silva, Sona Jain, Marilise Brittes Rott, Ricardo Scher, Cristiane Bani Correa, Ana Andrea Teixeira Barbosa, Silvio Santana Dolabella

Summary: This study demonstrated the effect of nisin on A. castellanii trophozoites, causing cell-cycle arrest but allowing recovery of growth after 72 hours. These findings suggest the potential use of nisin as an adjuvant in the treatment of Acanthamoeba spp. infections.

Epidemiological and diagnostic aspects of feline leishmaniasis with emphasis on Brazil: a narrative review

Luiz F. J. Nascimento, Tatyane M. Cirilo, Dharliton S. Gomes, Ana Carolina A. Gomes, Victor F. S. Lima, R. Scher, S. Jain, Ricardo T. Fujiwara, Silvio S. Dolabella

Summary: Feline leishmaniasis (FeL) is now considered an emerging disease in Brazil, with an estimated prevalence of 8% and Leishmania infantum being the most prevalent species. Cats infected with Leishmania typically show skin lesions, mainly in the cephalic region, and diagnosis is primarily conducted through serological tools. However, further research is needed on the immune response and diagnostic methods for FeL in cats.

Why is oral-induced RNAi inefficient in Diatraea saccharalis? A possible role for DsREase and other nucleases

Manoely Abreu Reis, Daniel David Noriega, Gessica dos Santos Alves, Roberta Ramos Coelho, Maria Fatima Grossi-de-Sa, Jose Dijair Antonino

Summary: The efficiency of RNAi technology varies in insects. This study found that gene silencing can be achieved in the sugarcane borer through feeding with dsRNA, but effective knockdown requires high dosages and long exposure times. The activity of gut nucleases may be one of the factors contributing to the low efficiency of RNAi in this species.

Summary: Schistosomiasis is a neglected tropical disease mainly found in Brazil, where Kato-Katz (KK) thick smear is the recommended screening test. However, KK has limitations in sensitivity. Helmintex (HTX), a magnetic-based method, showed superior performance compared to KK in a moderate endemic area in northeastern Brazil. This study confirms the potential role of HTX in defining true-positive samples and certifying transmission interruption of schistosomiasis.

Summary: In this study, Bacillus thuringiensis (Bt) Cry toxins were screened for their activity against the sugarcane giant borer, Telchin licus licus. Cry1A (a, b, and c) and Cry2Aa showed high toxicity against the larvae of this insect, with Cry1Ac being the most potent. Molecular dynamics and docking analyses provided insights into the potential interactions between Cry1Ac and aminopeptidase N (APN) receptors. These findings contribute to the development of transgenic sugarcane plants resistant to this important agricultural pest.

3-Carene-loaded poloxamer micelles against Leishmania: Development, characterization and in vitro proof-of-concept

Audrey R. S. T. Silva, Amanda M. B. Costa, Sona Jain, Patricia Severino, Ricardo Scher, Rogeria S. Nunes, Eliana B. Souto, Silvio S. Dolabella

Summary: Leishmaniases are parasitic diseases that need cheaper, safer, and more effective treatment options. The anti-fungal compound 3carene (3CR) has potential as a treatment for Leishmania, but its hydrophobicity limits therapeutic uses. In this study, micellar systems made from poloxamers were developed to carry 3CR and enhance its activity against Leishmania braziliensis.

Adelson Ferreira Ramos Neto, Yvanna Louise Di Christine Oliveira, Luciana Maria de Oliveira, Roseli La Corte, Sona Jain, Divaldo Pereira de Lyra Jr, Ricardo Toshio Fujiwara, Silvio Santana Dolabella

Summary: This overview examines the influence of environmental factors, host-related risk factors, and control strategies on the prevalence of soil-transmitted helminthiasis (STH) in different regions of the world. The results showed that factors such as climate, temperature, soil moisture, precipitation, mass drug administration, lack of access to water and sanitation, and non-use of footwear are associated with the prevalence of STH. Socioeconomic factors, low educational level, and wearing shoes are universal factors related to prevalence, regardless of the location studied.
