Summary: The study found that psoriasis patients had impaired HDL function at baseline compared to controls, but short-term and especially intermediate-term therapy with biologics markedly reduced HDL cholesterol efflux capacity and rendered HDL pro-inflammatory. However, it increased paraoxonase and lecithin-cholesterol acyltransferase activities. All biologics caused similar changes in HDL composition, subclass distribution and cholesterol efflux capacity.

Summary: By using simulations, the authors of this study uncover the mechanism by which ATP-binding cassette transporter A1 (ABCA1) functions in extracting lipids from the outer face of the plasma membrane and facilitating the production of high density lipoprotein (HDL). Contrary to existing models, their findings suggest that ABCA1 removes lipids from the plasma membrane and forces them through its gateway and annulus into an elongated hydrophobic tunnel. This study provides important insights into the function of ABCA1 and presents a potential target for further research.

Summary: ATP-binding cassette transporter A1 (ABCA1) is a molecular defect in Tangier disease, characterized by the absence of high-density lipoprotein cholesterol (HDL-C) in the circulation and the accumulation of cholesterol in lymphoid tissues. Atherosclerosis is caused by the accumulation of cholesterol in arteries, while HDL-C levels are inversely associated with the presence of atherosclerotic cardiovascular disease (ASCVD). Strategies to increase ABCA1 expression and activity have been studied to reduce ASCVD, but no successful clinical trials have been conducted yet. This review discusses the current understanding, challenges, and future directions in ABCA1 research for developing therapies for ASCVD.

Summary: Homozygotes with loss-of-function mutations in ABCA1 develop Tangier disease, which is characterized by significantly reduced or absent blood high-density lipoprotein (HDL) cholesterol. ABCA1 was identified as a key player in HDL production based on studies involving ABCA1-deficient mice. While the activation of ABCA1 and promotion of cholesterol efflux could potentially lead to the development of anti-atherosclerotic drugs, it is challenging to consider simple ABCA1 activation or cholesterol efflux promotion as an effective strategy for intracellular cholesterol homeostasis. Currently, there is no evidence supporting the preventive effect of HDL-increasing drugs that enhance ABCA1 expression on atherosclerotic disease in humans. However, in situations where intracellular cholesterol homeostasis is disrupted by inflammation, aging, or metabolic abnormalities, restoring ABCA1 expression and cholesterol efflux may be a useful strategy.

Summary: This study investigated the mechanism of docosahexaenoic acid (DHA)-induced apoptosis mediated by mitophagy, using grass carp as an animal model. The results showed that inhibition of mitophagy alleviated apoptosis and eliminated the inhibition of lipid accumulation induced by DHA. Mechanistically, DHA induced mitophagy by activating the PPAR gamma-LC3-BNIP3 pathway. Inhibition of PPAR gamma decreased autophagy-related gene expression and prevented BNIP3/NIX-mediated mitophagy-induced apoptosis, thereby alleviating the inhibition of lipid accumulation.

Summary: This study aims to investigate the role of the gut microbiota-bile acid axis in regulating the diurnal rhythms of metabolic homeostasis and assess the impact of obesity on them. The results show that high fat diet feeding and Leptin gene deficiency disrupt the rhythmic patterns of insulin sensitivity and serum total cholesterol levels. The study provides compelling evidence for the association between diurnal rhythm of insulin sensitivity and gut microbiota-bile acid axis, and elucidates the deleterious effects of obesity on gut microbiome-bile acid metabolism.
