Summary: Ion-pairing reversed-phase liquid chromatography (IP-RPLC) is the reference separation technique for characterizing oligonucleotides (ONs) and their related impurities. This study aimed to understand the retention mechanism of ONs, evaluate the applicability of the linear solvent strength (LSS) retention model, and explore ultra-short columns for ONs separation. It was found that 5-35 mm column lengths were appropriate for most gradient separation conditions. Baseline separation of three model ONs mixtures was achieved in as little as 30 seconds on the 5 mm column. This proof-of-concept work paves the way for future investigations using more complex therapeutic ONs and their related impurities.

Summary: To address the challenges in isolating and analyzing Circulating Tumour Cells (CTCs), researchers developed a high throughput Static Droplet Microfluidic (SDM) device capable of isolating and classifying metabolically active CTCs in peripheral blood at single-cell resolution. The device could accurately measure lactate production of different cancer cells and differentiate them from leukocytes. It was also able to detect cancer cells spiked in healthy blood. Testing on preclinical mouse models demonstrated the ability of the SDM device to distinguish metabolically active CTCs. This research provides a simple and robust workflow for on-site metabolic screening of cancer cells.

Summary: This study demonstrates the feasibility of using hyperspectral image analysis in the visible and near-infrared range for primary screening of SARS-CoV-2. By applying spectral feature descriptors, partial least square-discriminant analysis, and artificial intelligence, information can be extracted from fluid samples and analyzed quantitatively and descriptively. The proposed technology is reagent-free, fast, scalable, and could significantly reduce the number of molecular tests required for COVID-19 mass screening, even in resource-limited settings.

Summary: This study evaluated the antimicrobial potential of Terminalia ferdinandiana extracts using a 96-well plate reader as a high throughput screening method. The UV-VIS spectra analysis allowed for the differentiation between live and dead bacteria based on the interactions with the extracts. Partial least squares discriminant analysis successfully classified samples according to the concentration of extract added to the culture.

Summary: Gene doping, an approach to improve athletic performance, is prohibited and we developed a dCas9-based high-throughput gene doping analysis for exogenous gene detection. This method utilizes two different dCas9 proteins for signal amplification and can detect target genes in whole blood samples. We propose that this method, HiGDA, is a fast, highly sensitive, and practical detection method for future doping in sports.

Summary: In this study, an experimental combinatorial approach based on high-throughput sample preparation, nanoindentation platform, and EDS mapping was proposed to rapidly screen the mechanical properties of ZrNi-based metallic glasses. The mechanical properties, including elastic modulus, hardness, strength, and plastic deformation ability, were quickly characterized using nanoindentation. The composition ranges with optimal mechanical properties were identified separately.

Summary: This study utilized infrared spectral imaging (IRSI) to investigate the expression and distribution changes of glypicans in different hair follicle compartments during various phases of the hair growth cycle, providing a new perspective on hair shaft growth. The results demonstrate the capability of IRSI to simultaneously reveal the location of proteins, PGs, GAGs, and sulfated GAGs in hair follicles in a reagent- and label-free manner.
