Summary: A series of hydrazonobenzenesulfonamides were designed, synthesized and evaluated for their inhibitory activity against human carbonic anhydrase (hCA) isoforms. The synthesized compounds showed inhibitory activity at low nanomolar levels against hCA I, II, IX and XII, with selectivity against hCA II isoform, as well as hCA IX and XII isoforms. Docking studies were conducted to understand the activity and selectivity of the most potent and selective hydrazones towards different CA isoforms.

Summary: Rhodanine and its derivatives are a valuable class of heterocycles with diverse biological properties, including anticancer, antibacterial, and anti-mycobacterial activities. In this study, new Rhodanine derivatives were synthesized and evaluated for their inhibitory activity against carbonic anhydrase isoforms. The compounds showed good inhibitory activity against cytosolic isoform hCA II and tumor-associated hCA IX. The mechanism of action was supported by molecular docking studies.

Summary: Carbonic Anhydrases (CAs) are important targets for treating various disease conditions, but the difficulty lies in finding potential drugs due to their ubiquitous localization and high homology. By synthesizing new bicyclic tetrahydroindazoles and conducting biological evaluations and theoretical studies, a new platform for the discovery of isotype selective CA inhibitors has been identified.

Compound 1a exhibited strong inhibitory activity and showed potential as an anticancer agent, providing valuable insights for further research.

Summary: A new class of novel sulfonamides with aminosaccharide tails were designed and synthesized, showing effective inhibition against three carbonic anhydrase isoforms, particularly hCA II. The compounds exhibited good water solubility, neutral pH values, and lower corneal cytotoxicity compared to acetazolamide, making them potential candidates for topical antiglaucoma drugs. Molecular docking studies demonstrated the crucial role of interactions between the aminosaccharide fragment and the hydrophilic subpocket of hCA II in enhancing CA inhibitory activity. Compound 8d was identified as a promising lead compound for further development.

Summary: The carbonic anhydrase (CA) family not only catalyzes the reversible hydration of CO2 to bicarbonate, but also possesses esterase and phosphatase activity. Recent studies have shown that CA II can convert inorganic nitrite to nitric oxide and nitrous anhydride, potentially representing a bioactivation of nitrite. The key role of L-cysteine in this bioactivation process is supported by new experimental data.

Summary: Acetylcholinesterase (AChE) and carbonic anhydrase I (CA-I) and II (CA-II) are important metabolic enzymes. This study synthesized four benzimidazole acetamide derivatives and investigated their inhibition effects on AChE, hCA-I, and hCA-II. The results showed that these compounds exhibited strong inhibition effects on AChE and carbonic anhydrase.
