The genetic diversity and population genetic structure of Culter alburnus were investigated using mitochondrial cytochrome c oxidase subunit I (COI) and ribosomal 16S subunit (16S rRNA) gene sequences. A total of 89 individuals from four localities were included in the analysis. Overall, 12 polymorphic sites were observed and 10 haplotypes were defined. The C. alburnus populations were characterized by high haplotype diversity (0.587 +/- 0.047) and low nucleotide diversity (0.00197 +/- 0.00073). Pairwise fixation index (F-ST) analysis indicated significant genetic differentiation among different populations. The hierarchical analysis of molecular variance (AMOVA) analysis also showed significant genetic divergence (phi(ST) = 0.3792, P < 0.01) among these populations. The present results suggest that subdivisions exist among four C alburnus populations, and should be considered as different management unit for effective conservation and management purposes. This study provides new information for genetic assessment and will be crucial for establishing fisheries management and strategies for this species. (C) 2013 Elsevier Ltd. All rights reserved.

Huai Yang, Zhongtian Xu, Baoying Guo, Xiaolin Zhang, Zhi Liao, Pengzhi Qi, Xiaojun Yan

Summary: The study revealed a sophisticated miRNA-mRNA regulation network in M. coruscus in response to V. alginolyticus challenge, linking immune-related signaling pathways with specific miRNAs and genes. It also dissected the interactional relationship between certain miRNAs and the TLR pathway, predicting negative regulation of TLRs and associated genes by novel miRNAs. Additionally, a laboratory experiment validated the interactional relationship between novel miR-145 and TLR2.

Micro-polyethylene particles reduce the toxicity of nano zinc oxide in marine microalgae by adsorption*

Jiji Li, Shuai Mao, Yingying Ye, Jiayin Lu, Fei Jing, Yahong Guo, Hongxia Liu, Ping Wang, Wei Ma, Pengzhi Qi, Jialang Zheng, Chengkai Qu

Summary: Microplastics adsorb contaminants and can mitigate the toxic effects of certain pollutants on microorganisms, as demonstrated by combined-exposure assays showing that polyethylene microplastics can alleviate the toxicity of nano zinc oxide on marine microalgal growth. This interaction may be achieved through zinc oxide adsorption onto the polyethylene microplastic surface.

A novel molluscan TLR molecule engaged in inflammatory response through MyD88 adapter recruitment

Pengzhi Qi, Yashu Wu, Zhongqi Gu, Hongfei Li, Jiji Li, Baoying Guo, Zhi Liao, Xiaojun Yan

Summary: This study identified a novel TLR isoform called McTLRlike1 in the mollusk Mytilus coruscus. McTLR-like1 was highly expressed in immune-related tissues and its levels increased when challenged with V. alginolyticus. It was found that McTLR-like1 activated NF-kappa B and influenced the expression and phosphorylation of its subunit P65. Silencing McTLR-like1 led to downregulation of pro-inflammatory cytokines and increased resistance to V. alginolyticus challenge. McTLR-like1 also showed affinity with McMyD88a. These findings contribute to our understanding of TLR signaling in mollusks and provide insights into TLR evolution.

Summary: A novel AMP family called myticofensins, similar to arthropod defensins, was identified in Mytilus coruscus. These myticofensins showed high expression levels in immune-related tissues and their expression could be significantly increased under microbial challenges. Different myticofensins may have different immune functions in different tissues.

Proteomic analysis of the spineless cuttlefish Sepiella japonica: Exploratory analyses on the phenomenon of death after spawning

Hongfei Li, Shuangrui Lin, Pengzhi Qi, Jingtian Wang, Meihua Fan, Hongwei Huang, Zhenyu Dong, Kaida Xu, Baoying Guo

Summary: In this study, a proteomic data set was generated to investigate the physiological events involving death after spawning in S. japonica. Differential expression proteins (DEPs) were identified using a proteomic-based approach, and their functions were found to be mainly related to gene translation and signal transduction. The study also discovered aging-related DEPs in addition to those involved in signal transduction and energy metabolism. Overall, this study provides new insights into the phenomenon of death after spawning in S. japonica at the protein level.

Distinguish the toxic differentiations between acute exposure of micro- and nano-plastics on bivalves: An integrated study based on transcriptomic sequencing

Pengzhi Qi, Longmei Qiu, Dan Feng, Zhongqi Gu, Baoying Guo, Xiaojun Yan

Summary: Plastic pollution is a severe marine environmental issue. This study investigated the toxic effects of acute exposure to different sizes of plastic particles on mussels using various techniques. The results showed that mussels efficiently ingested nanoplastics and experienced a severe inflammatory response, while microplastics caused a slight inflammatory response. Biochemical analysis revealed up-regulation of biomarkers after plastic particle exposure. Nanoplastics induced more ROS production and higher T-AOC level than microplastics. Transcriptomic sequencing indicated that differentially expressed genes were mostly involved in stress and immune response pathways. Acute exposure to nanoplastics was found to be more toxic than microplastics. Microplastics exposure may impair olfactory function and cause neurotoxicity to mussels. These findings provide new insights into the ecotoxicological mechanisms of plastic particle exposure.

Exploring the Role of a Novel Interleukin-17 Homolog from Invertebrate Marine Mussel Mytilus coruscus in Innate Immune Response: Is Negative Regulation by Mc-Novel_miR_145 the Key?

Xinglu Chen, Longmei Qiu, Xirui Si, Xiaolin Zhang, Baoying Guo, Zhi Liao, Xiaojun Yan, Pengzhi Qi

Summary: This study investigated the association between Mc-novel_miR_145 and IL-17 homolog (McIL-17-3) and their immunomodulatory effects in Mytilus coruscus. The results showed that McIL-17-3 was highly expressed in immune-associated tissues and responded to bacterial challenges. McIL-17-3 could activate downstream NF-kappa b and was negatively regulated by Mc-novel_miR_145. Moreover, Mc-novel_miR_145 could alleviate LPS-induced apoptosis by negatively regulating McIL-17-3.

An Interleukin-17 Isoform from Thick Shell Mussel Mytilus coruscus Serves as a Mediator of Inflammatory Response

Jiemei Zhao, Zhenyu Dong, Li Zhu, Weihua Song, Pengzhi Qi

Summary: The newly identified member of the IL17 family, McIL17-1, plays an important role in the innate immune response to bacterial infection in M. coruscus. It is expressed in various tissues with high levels in gills, hemocytes, and digestive glands. McIL17-1 is upregulated after stimulation with different pathogen associated molecular patterns (PAMPs) and has a cytoplasmic localization in HEK293T cells. Knockdown of McIL17-1 reduces mortality rate in response to Vibrio alginolyticus infection, while overexpression increases mortality rate.
