The study also showed an up-regulation of glutamate transporter 1 in the intoxicated liver, contrasting with a time-dependent reduction in glutamine synthetase, indicating a disruption of glutamate transporters expression in the liver-brain axis under acute CCl4 insult.

Summary: This study reveals that Cav1 deficiency in mice provides protection against acute liver injury, reducing hepatocyte degeneration, necrosis, and apoptosis as well as lowering ALT levels. Cav1 is also crucial for the polarization of hepatic macrophages, with its deficiency inhibiting the polarization towards the M1 phenotype. These findings suggest that Cav1 expressed in macrophages could be a potential therapeutic target for acute liver injury.

Summary: The enzyme asparagine synthetase (ASNS) is responsible for the de novo synthesis of the nonessential amino acid asparagine. The expression of ASNS in pericentral hepatocytes, specialized liver cells in xenobiotic detoxification, is significantly increased in models of acute liver injury. Mice with hepatocyte-specific Asns deletion are more susceptible to pericentral liver damage, which can be reversed by intravenous administration of asparagine. The upregulation of ASNS in response to stress is mediated by the nuclear receptor, liver receptor homolog 1 (LRH-1; NR5A2) through an ATF4-independent, noncanonical pathway.

Summary: The mechanism of accelerated aging in thyrotoxicosis mice was investigated in this study. Excess thyroxine led to cardiac hypertrophy, decreased hepatic synthetic capacity, and decreased hepatic and renal metabolic capacity. The main cause of organ senescence was identified as proteotoxicity, which resulted from intensified oxidative phosphorylation, excessive ribosome production, and impaired splicing and ubiquitin proteasome system function. This research provides a convenient and suitable animal model for studying aging mechanisms and antiaging drugs.

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

Summary: This study explored the inhibitory effects of gallium curcumin, indium curcumin, and vanadyl curcumin on the amyloid fibrillation of hen egg white lysozyme, as well as the binding interactions of these metal complexes with the enzyme. The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.
