Summary: Proteinase-activated receptors (PARs) in prostate cancer cells regulate tumor growth and metastasis through autocrine signaling. In this study, PAR1 and PAR2 were found to be functional in PC3 cells, while PAR4 was not expressed. The secretion of proteolytic enzymes by PC3 cells triggers cleavage of PARs, leading to autocrine signaling and the regulation of genes related to prostate cancer. PAR1 deficiency promotes cell migration and suppresses cell proliferation, while PAR2 deficiency has the opposite effects.

Summary: Endometrial mesenchymal stem cells (eMSCs) have the ability to differentiate into different cell types and are influenced by mechanical forces in their microenvironment. This study identified Piezo1 proteins as mechanosensitive channels in the plasma membrane of eMSCs, which transduce mechanical stimuli into intracellular signaling pathways. The influx of Ca2+ triggered by Piezo1 activity is regulated by store-operated Ca2+ entry (SOCE) through the ORAI1 channel and STIM1/STIM2 Ca2+ sensors. Activation of Piezo1 or SOCE does not cause cytotoxicity in eMSCs but affects their migratory capacity and cell proliferation. This finding suggests that Piezo1 and SOCE play important roles in the regulation of intracellular calcium, which influences the migratory activity and regenerative potential of eMSCs.

Summary: The study demonstrates that NBCe1 plays a crucial role in regulating acid extrusion in prostate cancer cells, and inhibiting or abolishing this transporter can decrease cancer cell proliferation. Knockdown or inhibition of NBCe1 reduces cell viability and increases cell death, and also inhibits the formation of cell spheres in 3D cultures. Additionally, immunohistochemistry reveals the localization of NBCe1 in glandular epithelial cells in prostate tissue and its robust expression in acinar and duct adenocarcinoma.

Summary: The study aimed to evaluate the activity of a triazaspirane as a migration and invasion inhibitor in PC3 prostatic tumor cells through negative regulation of the FAK/Src signal transduction pathway and decreased secretion of metalloproteinases 2 and 9. Molecular docking analysis showed interactions in regions of interest of the FAK and Src proteins. Biological activity assays demonstrated inhibitory effects on cell migration and invasion, suppression of metalloproteinase secretion, and decreased expression of p-FAK and p-Src proteins in treated PC3 cells. Triazaspirane-type molecules have important inhibitory effects on metastasis-associated mechanisms in PC3 tumor cells.

Summary: The study showed that suppression of miR-21 significantly reduced cell viability of PC3/DOX, downregulated P-gp expression and activity in DOX-resistant cells, and reversed MDR by increasing the intracellular accumulation of DOX in PC3/DOX cells. Additionally, miR-21 repression led to the upregulation of PTEN and subsequently lower expression of P-gp, enhancing the apoptosis rate of PC3/DOX cells. The findings contribute to the understanding of the role of miR-21 in reversing MDR in prostate cancer.

The development of defects in genetically modified animal models differs notably from human diseases at the molecular level.

Summary: Prostate cancer is the second most frequent and sixth most fatal cancer in men worldwide. Recent studies have suggested the potential participation of exosomes in the progression of prostate cancer. This study isolated exosomes from the serum of prostate cancer patients and studied their effects on tumor aggressiveness in two prostate cancer cell lines. The results showed that serum exosomes from prostate cancer patients increased migration of PC3 cells and neuroendocrine differentiation of LNCaP-FGC cells.
