Summary: This study identified rare pathogenic genetic variants linked to Chagas disease cardiomyopathy in multiple nuclear families, mainly involving mitochondrial or inflammation-related genes. The findings suggest a genetic basis for mitochondrial dysfunction and inflammation in CCC, potentially affecting myocardial susceptibility to IFN-gamma-induced damage and contributing to the cardiomyopathy phenotype in Chagas disease and possibly other inflammatory cardiomyopathies.

Furthermore, miRNAs hsa-miR-16 and hsa-miR-200a-3p were identified as potential biomarkers for T1D progression.

Using machine learning algorithms, predictions in a test cohort achieved an accuracy of over 95%. Summary: In this study, the genetic and epigenetic deregulations associated with moderate and severe stages of Chagas disease were investigated using bulk RNA-seq and DNA methylation analysis. The results revealed differentially expressed transcripts and regulatory differentially methylated regions in the heart tissue of CCC patients. Transcription factors binding sites affected by DNA methylation were also identified. The study identified biological processes affected by severe CCC and highlighted the importance of ion transport and the nervous system in the development of the disease.

Summary: This study performed miRNA profiling of patients before coronary artery bypass grafting (CABG) surgery to identify miRNAs that could predict Vasoplegic syndrome (VS). Six miRNAs were increased and two were decreased in patients who developed VS compared to those who did not. The combination of hsa-miR-30d-5p and hsa-miR-770-5p showed high accuracy in predicting VS.

Uncoupling protein 2-mediated metabolic adaptations define cardiac cell function in the heart during transition from young to old age

Summary: This study reveals a novel role for uncoupling protein 2 (UCP2) in regulating functional properties of cardiac tissue-derived stem-like cells (CTSCs). UCP2, highly expressed in neonatal CTSCs, is linked to glycolysis, proliferation, and survival, but its reduction with age shifts cellular metabolism to oxidative phosphorylation, affecting proliferation and survival in aged CTSCs. Loss of UCP2 in neonatal CTSCs leads to reduced glycolysis, proliferation, and survival, with significant alterations in mitochondrial genes identified by RNA sequencing and bioinformatic analysis.
