Endoglin (CD 105, TGF-beta receptor III) is a homodimeric transmembrane glycoprotein that plays a regulatory role in TGF-beta signaling. Its functional role in the context of atherosclerosis has yet to be defined and should be stated here. Therefore, we focused on the role of endoglin in atherosclerosis in both humans and experimental animals. Endoglin expression was demonstrated in atherosclerotic vessels predominantly in endothelial cells and smooth muscle cells in various types of blood vessels in mice and humans, suggesting its participation in atherogenesis. Endoglin expression was also related to the expression of eNOS in endothelium, repair of the vessel wall, plaque neoangiogenesis, production of collagen and stabilization of atherosclerotic lesions. In addition, increased levels of soluble endoglin were associated with hypercholesterolemia, atherosclerosis, acute myocardial infarction and were related to inhibition of TGF-beta signaling in the vessel wall. Moreover, soluble endoglin levels were significantly lowered after a series of extracorporeal eliminations in patients with familial hypercholesterolemia. Additionally, statin treatment decreased levels of soluble endoglin and increased its expression in aorta, which was related to reduced atherosclerosis in mice. In conclusion, we propose that measurement of soluble endoglin might give information about progression of the atherosclerotic process or the efficacy of therapeutic interventions, which is the task that must be answered in clinical trials. Monitoring of up to 15 years effects of lipoprotein apheresis on lipids, biomarkers of inflammation, and soluble endoglin in familial hypercholesterolemia patients

J. Visek, M. Blaha, V Blaha, M. Lasticova, M. Lanska, C. Andrys, J. Duintjer Tebbens, Ivone Cristina Igreja e Sa, K. Tripska, M. Vicen, I Najmanova, P. Nachtigal

Summary: The study analyzed data from FH patients undergoing lipoprotein apheresis (LA) treatment over 15 years, showing that long-term LA treatment significantly reduced cholesterol levels, improved endothelial dysfunction, and decreased levels of inflammation biomarkers in FH patients.

Metformin impairs bile acid homeostasis in ethinylestradiol-induced cholestasis in mice

Fatemeh Alaei Faradonbeh, Ivone Igreja Sa, Hana Lastuvkova, Jolana Cermanova, Milos Hroch, Hana Faistova, Jaroslav Mokry, Zuzana Nova, Martin Uher, Petr Nachtigal, Petr Pavek, Stanislav Micuda

Summary: Metformin was found to stimulate bile secretion in healthy mice by increasing liver bile acid synthesis and intestinal bile acid reabsorption. However, when used in combination with estrogen, metformin exacerbated bile secretion impairment and led to a significant increase in plasma bile acid concentrations.

Summary: Atorvastatin modulates bile acid metabolomics significantly by altering their intestinal processing and liver synthesis in control and NASH mice, improving liver conditions and reducing steatosis and inflammation in NASH animals.

Study of the impact of cultivation conditions and peg surface modification on the in vitro biofilm formation of Staphylococcus aureus and Staphylococcus epidermidis in a system analogous to the Calgary biofilm device

Adela Diepoltova, Klara Konecna, Ondrej Jand'ourek, Petr Nachtigal

Summary: The study found that under specific conditions, the supplementation of tryptic soy broth with HP and using the shaking mode significantly promoted biofilm formation on pegs for Staphylococcus aureus. However, Staphylococcus epidermidis did not form biofilms on pegs under the same conditions. Preconditioning peg surfaces with FBS and HP led to a significant increase in biofilm biomass production for Staphylococcus epidermidis.

Monoclonal anti-endoglin antibody TRC105 (carotuximab) prevents hypercholesterolemia and hyperglycemia-induced endothelial dysfunction in human aortic endothelial cells

Summary: This study found that blocking the endoglin receptor using TRC105 can counteract endothelial dysfunction induced by high cholesterol and high glucose, suggesting that endoglin may be a potential therapeutic target for disorders associated with elevated cholesterol and glucose levels.

Labetalol and soluble endoglin aggravate bile acid retention in mice with ethinylestradiol-induced cholestasis

Ivone Cristina Igreja Sa, Katarina Tripska, Fatemeh Alaei Faradonbeh, Milos Hroch, Hana Lastuvkova, Jolana Schreiberova, Marian Kacerovsky, Miguel Pericacho, Petr Nachtigal, Stanislav Micuda

Summary: Labetalol exacerbates the accumulation of bile acids (BA) in estrogen-induced cholestasis and soluble endoglin (sEng) aggravates this retention. Increased sEng levels might explain the coincidence of intrahepatic cholestasis of pregnancy (ICP) with preeclampsia. Monitoring of BA in pregnant women with preeclampsia and labetalol therapy is recommended.

Summary: The study demonstrated that long-term exposure to high levels of sEng during aging can aggravate vascular function, leading to alterations in vasoconstriction properties and Eng signaling pathway. sEng may be considered a potential risk factor for the development of vascular dysfunction during aging.
