Summary: The study demonstrates that steroid substrates have different effects on the affinity and stability of CYP11s-Adx complexes, with cholesterol increasing the formation rate of CYP11A1 - Adx complex and decreasing the stability of CYP11B1 - Adx and CYP11B2 - Adx complexes. This differentiation in protein-protein interactions within the mitochondrial pool of CYPs reveals the potential impact of substrate regulation on overall steroid hormones production. The experimental data provide further insights into the redox chains involved in steroidogenesis.

Summary: Pediatric adrenocortical hyperplasias are rare conditions characterized by Cushing syndrome and genetic defects affecting the adrenal cortex. Most cases are caused by defects in the cyclic AMP/protein kinase A (cAMP/PKA) pathway, while a few cases have unidentified genetic defects. Congenital adrenal hyperplasia, mainly due to steroidogenic enzyme deficiencies, is another cause of adrenal hyperplasia in childhood.

Summary: The study determined the furan-containing components (FCCs) in Cortex Dictamni extract and found that four FCCs (RUT, LIM, OBA, and FRA) were bioactivated in vitro and in vivo. Among them, FRA displayed significant hepatotoxicity both in vitro and in vivo. In addition, FRA caused the depletion of hepatic glutathione (GSH) and the formation of GSH conjugates the most. The intrinsic clearance rate of the four FCCs followed the order: FRA>>OBA>LIM>RUT.

Summary: Variable expression of steroidogenic enzymes in adrenal adenoma underlies the clinical heterogeneity of PA and is associated with treatment outcomes. CYP11B1 expression was positively correlated with the severity of hypercortisolemia, and CYP11B2 was positively correlated with that of hyperaldosteronism. Patients with absent clinical improvement after adrenalectomy had lower CYP11B2 expression than those with complete success.

Summary: A LC-MS method was developed for the analysis of 29 cortical steroids and 6 medullary amines in a single run. This method improved the validity of bioanalysis while maintaining sample homogeneity. The method was validated and showed satisfactory lower limits of quantification, recovery rates, and precision and accuracy. It was successfully applied to analyze serum samples from patients with adrenal diseases and characterized the metabolic features of different adrenal tumors.

Summary: This study evaluates the discriminatory capacity of three-dimensional volumetry on computed tomography (CT) to identify possible autonomous cortisol excess (ACS) in patients with adrenal adenomas. The results show that CT volumetry of adrenal adenomas and contralateral adrenal glands has a high ability to identify ACS. The combination of CT volumetry and biochemical testing can substantially improve the identification of ACS.
