Anticholinergic drugs and forebrain magnetic resonance imaging changes in cognitively normal people and those with mild cognitive impairment

Dewen Meng, Ali-Reza Mohammadi-Nejad, Stamatios N. Sotiropoulos, Dorothee P. Auer

Summary: The use of anticholinergic medication is associated with cognitive decline and structural and functional impairment of the cholinergic nucleus basalis of Meynert. The study found that individuals on long-term anticholinergic medication have lower gray matter density and reduced functional connectivity in the nucleus basalis of Meynert, leading to cognitive impairment.

Added value of magnetic resonance spectroscopy for diagnosing childhood cerebellar tumours

Nigel P. Davies, Heather E. L. Rose, Karen A. Manias, Kal Natarajan, Laurence J. Abernethy, Adam Oates, Umair Janjua, Paul Davies, Lesley MacPherson, Theodoros N. Arvanitis, Andrew C. Peet

Summary: In a prospective study, the diagnostic accuracy of a classifier based on MRS for childhood cerebellar tumors was evaluated, with an overall accuracy of 91%. Retrospective analysis showed that the combination of MRS with a decision support tool significantly increased radiologists' certainty in correct diagnosis. This suggests that MRS can aid in noninvasive diagnosis of posterior fossa tumors in children and that decision support tools can improve MRS interpretation.

Efficacy and Safety of Trametinib Monotherapy or in Combination With Dabrafenib in Pediatric BRAF V600-Mutant Low-Grade Glioma

Eric Bouffet, Birgit Geoerger, Christopher Moertel, James A. Whitlock, Isabelle Aerts, Darren Hargrave, Lisa Osterloh, Eugene Tan, Jeea Choi, Mark Russo, Elizabeth Fox

Summary: This study establishes pediatric dosing and pharmacokinetics, as well as efficacy and safety, of trametinib with or without dabrafenib in children with BRAF V600-mutant LGG. The recommended phase II dose of trametinib, with or without dabrafenib, was determined as 0.032 mg/kg or 0.025 mg/kg. Both monotherapy and combination therapy showed efficacy and tolerability in treating BRAF V600-mutant LGG.

Predictors of long-term disability in multiple sclerosis patients using routine magnetic resonance imaging data: A 15-year retrospective study

Amjad Altokhis, Abdulmajeed Alotaibi, Paul Morgan, Radu Tanasescu, Nikos Evangelou

Summary: MRI measures such as inter-caudate diameter (ICD) and third ventricular width (TVW) are correlated with long-term disability in multiple sclerosis (MS) patients. They show potential as simple and fast predictors of disease progression.

Applying machine learning classifiers to automate quality assessment of paediatric dynamic susceptibility contrast (DSC-) MRI data

Stephanie B. Withey, Yu Sun, Jan Novak, Lesley Macpherson, Laurence Abernethy, Barry Pizer, Richard Grundy, Paul S. Morgan, Tim Jaspan, Simon Bailey, Dipayan Mitra, Dorothee P. Auer, Shivaram Avula, Theodoros N. Arvanitis, Andrew Peet

Summary: The objective of this study was to investigate the performance of qualitative review (QR) in assessing the quality of dynamic susceptibility contrast (DSC-) MRI data in pediatric normal brain, and to develop an automated alternative to QR. Two reviewers independently evaluated 1027 signal-time courses, and their results showed 7% disagreement with a Cohen's kappa value of 0.83. The study determined data quality thresholds for various measures, trained machine learning classifiers, and found that SDNR provided the best performance.

Clinical Decision Support Systems for Brain Tumour Diagnosis and Prognosis: A Systematic Review

Teesta Mukherjee, Omid Pournik, Sarah N. Lim Choi N. Keung, Theodoros N. N. Arvanitis

Summary: Brain tumours are abnormal growths of cells in the human brain. Clinical decision support systems (CDSSs) have played an important role in the diagnosis and treatment of brain tumours. This study systematically identifies different types of CDSSs used in brain tumour diagnosis and prognosis through medical imaging.

A novel diagnostic method for a rare fungus: FcMBL facilitates Wickerhamomyces anomalus identification in an immunocompromised neonate

Conor J. Higgins, Kerry-Anne Kite, Nigel Klein, Michael Super, Michael T. Mccurdy, Darren Hargrave

Summary: Fungemia has a negative impact on patient outcomes, but current diagnostic methods lack sensitivity in identifying emerging rare fungal infections. This study reports a case of Wickerhamomyces anomalus infection in an immunocompromised neonate, where FcMBL bead-based matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS) identified the species approximately 30 hours before standard pathogen identification methods. The use of FcMBL bead-based MALDI-TOF MS may improve the speed and accuracy of identifying rare pathogens, leading to more effective treatment.

Platform-agnostic electrochemical sensing app and companion potentiostat

Vijayalaxmi Manoharan, Rui Rodrigues, Sara Sadati, Marcus J. Swann, Neville Freeman, Bowen Du, Ender Yildirim, Ugur Tamer, Theodoros N. Arvanitis, Dmitry Isakov, Ali Asadipour, Jerome Charmet

Summary: Researchers have developed an application called PocketEC, which can interface with multiple potentiostat ICs. Through Bluetooth technology, PocketEC can be connected to evaluation boards, open-source potentiostats, and custom-built potentiostats. The versatility of PocketEC was demonstrated by comparing its performance with standard benchtop potentiostats using various experiments. The application, along with its companion devices, can be used for a variety of wearable or portable electrochemical sensing applications.

Summary: This study investigated the impact of the WHO classification on pediatric high-grade gliomas. It found differences between neuro-oncologists and neuropathologists in their perception of molecularly defined tumor types and the changes made in the classification. While improvements were perceived in the 2021 edition, there are still relevant issues that need to be addressed, emphasizing the need for collaboration between pathologists and oncologists.
