Summary: Solar radiation can lead to various negative effects on the skin, such as sunburn, inflammation, and DNA damage. One potential approach to photoprotection is by targeting the transcription factor Nrf2, and obacunone has been identified as a potent Nrf2 agonist. This study aims to evaluate the relevance of full-thickness skin models in photoprotection studies and explore the photoprotective effects of obacunone in human keratinocytes.

Summary: In this study, THP-1-derived iDCs were integrated into a human skin model as central mediators of the immune reaction. The researchers found that different treatments of iDCs resulted in changes in proteins and cytokines related to sensitization and anti-sensitization. This provides a new approach for studying sensitizing agents and potential drugs for suppressing sensitization.

Summary: This study evaluated the function and preclinical validation of human dendritic cell (DC) dexosomes, small DC-secreted vesicles that contain immune signals, for cancer vaccination. The researchers characterized a potential dexosome model using immature and mature DCs derived from a cell line called MUTZ3, and found that one type of MUTZ3-derived DC dexosomes exhibited potential immunogenicity and higher antigen presentation potency, making them potential resources for cancer immunotherapy.

Summary: Research has shown that the epigenetic regulator miR-23a is crucial for LC antigen uptake and bone marrow-derived LC repopulation during inflammatory conditions. miR-23a regulates LC phagocytosis and repopulation by targeting molecules involved in efferocytosis, endocytosis, and proapoptotic pathways. This study reveals the context-dependent regulatory role of miR-23a in LCs, providing insights into the TGF-b- and PU.1-mediated regulation during steady-state and inflammation-induced repopulation.

Summary: In this study, increased expression and activity of TG2 were observed in systemic sclerosis (SSc) patients. Inhibition of TG2 in dermal fibroblasts resulted in reduced fibrosis, suggesting that targeting TG2 may be a potential antifibrotic approach in SSc.

Summary: Tissue engineering is an important and widely applicable field that offers alternatives to animal models and enables tissue regeneration and organ transplantation. 3D bioprinting stands out for its flexibility and reproducibility. A study introduces an integrated fluorescent light sheet bioprinting and imaging system, which allows real-time monitoring of material crosslinking and cell activity, offering high speed, high resolution, and real-time characterization capabilities.

Summary: This study investigates the regulatory functions of mast cells (MCs) in the skin, focusing on their interaction with regulatory T lymphocytes (Tregs) and Langerhans cells (LCs) during passive cutaneous anaphylaxis (PCA) responses. The researchers found that Tregs increased their contact time with MCs during PCA, suggesting an anti-inflammatory role in preventing further MC activation.

Summary: This study evaluated the benefits of full-thickness skin grafts in the reconstruction of midface defects after maxillectomy. The results showed that full-thickness skin grafts contributed to patient's convalescence and reduced postoperative pain, making them a good alternative for patients who are not eligible for microsurgical treatment.

Summary: In this study, a simple and fast method was developed to directly process adipose tissue into microfragmented adipose extracellular matrix (mFAECM) for 3D bioprinting. The mFAECM composite demonstrated biocompatibility, printability, and fidelity in vitro, and accelerated wound healing in a full-thickness skin defect model in nude mice.

Summary: In skin research, traditional 2D cell models are not sufficient in mimicking physiological properties, so there is a need for new 3D skin models. This study designed cell culture inserts fabricated with 3D printers for generating full-thickness skin models. The improved design of the inserts allows for easier and more efficient handling.

Summary: This study designed an adhesive antibacterial hydrogel dressing based on dynamic host-guest interaction, inspired by mussel. The hydrogel showed strong adhesion and on-demand removal, along with rapid self-healing properties. It also exhibited excellent performance in promoting wound healing and reducing scar formation.

Summary: LCs in the epidermis contain genes that contribute to antimicrobial responses, including chemokines such as CCL1, CCL17, CCL19, CCL2, CCL22, CXCL14, and CXCL2, which mediate both antimicrobial and inflammatory responses. These genes encode proteins with broad antibacterial activity, including against Staphylococcus aureus, the leading cause of skin infections.

Summary: Photobiomodulation (PBM) is a valuable tool in dermatology for treating inflammatory skin conditions using low levels of light, but the physiological pathways responsible for its anti-inflammatory effect have not been well defined. This study found that PBM activated the Nrf2 pathway to control inflammation and played a key role in reducing inflammatory response. The findings support the role of PBM in skin immunomodulation and suggest it as a promising alternative or complementary therapeutic approach for skin-related inflammatory diseases.
