Comet Assay on Daphnia magna in eco-genotoxicity testing

Detection of potentially hazardous compounds in water bodies is a priority in environmental risk assessment. For the evaluation and monitoring of water quality, a series of methodologies may be applied. Among them, the worldwide used toxicity tests with organisms of the genus Daphnia is one of the most powerful. In recent years, some attempts were made to utilize Daphnia magna in genotoxicity testing as many of the new environmental contaminants are described as DNA-damaging agents in aquatic organisms. The aim of this research was to develop a highly standardized protocol of the Comet Assay adapted for D. magna, especially regarding the isolation of cells derived from the same tissue (haemolymph) from newborn organisms exposed in vivo. Several methods for haemolymph extraction and different Comet Assay parameters were compared. Electrophoretic conditions were adapted in order to obtain minimum DNA migration in cells derived from untreated organisms and, at the same time, maximum sensitivity in specimens treated with known genotoxicants (CdCl2 and H2O2). Additional tests were performed to investigate if life-history traits of the cladoceran (such as the age of adult organisms that provide newborns, the clutch size of origin, the number of generations reared in standard conditions) and the water composition as well, might influence the response of the assay. This study confirms the potential application of the Comet Assay in D. magna for assessing genotoxic loads in aqueous solution. The newly developed protocol could integrate the acute toxicity bioassay, thus expanding the possibility of using this model species in freshwater monitoring (waters, sediment and soil elutriates) and is in line with the spirit of the EU Water Framework Directive in reducing the number of bioassays that involve medium-sized species. In Vitro Sprouted Plantlets of Citrullus colocynthis (L.) Schrad Shown to Possess Interesting Levels of Cucurbitacins and Other Bioactives against Pathogenic Fungi

Summary: Cucurbitacins, bioactive compounds found in Cucurbitaceae plants, have immense pharmacological potential. Citrullus colocynthis has been widely used in folk medicine due to its high content of cucurbitacins. However, the cultivation conditions and seed germination of C. colocynthis limit the exploitation of its phytochemical potential. This study evaluated axenic sprouted plantlets cultivated under laboratory conditions and found that they contain cucurbitacins and other bioactive compounds, exhibit antioxidant potential, and show activity against pathogenic fungi. These findings support the feasibility of scaling up the cultivation of C. colocynthis as a valuable natural resource for pharmaceutical compounds.

Post-Translational Modifications of Histone Variants in the Absence and Presence of a Methionine-Depleting Enzyme in Normal and Cancer Cells

Summary: Cancer cells have a higher requirement for methionine compared to healthy cells, but the mechanism behind their higher sensitivity to methionine depletion is still unclear. Methionine plays a crucial role in the epigenetic reprogramming of cells and the modification of histone variants. Mass-spectrometry analysis was used to characterize the post-translational modifications of histone variants in normal and cancer cells in the presence and absence of methionine gamma-lyase (MGL). The results revealed a complex pattern of modifications and significant differences between normal and cancer cells, providing insight into the molecular mechanisms triggered by methionine depletion and its potential therapeutic application with MGL.

Modulation of Transcription Profile Induced by Antiproliferative Thiosemicarbazone Metal Complexes in U937 Cancer Cells

Summary: Since the discovery of cisplatin, the search for metal-based compounds with therapeutic potential has been a challenge for the scientific community. Thiosemicarbazones and their metal derivatives, such as the three metal thiosemicarbazones [Ni(tcitr)(2)], [Pt(tcitr)(2)], and [Cu(tcitr)(2)] investigated in this study, show promise as anticancer agents with high selectivity and low toxicity. The study deepened the understanding of the molecular action mechanism of these compounds and demonstrated their potential as candidate antiproliferative thiosemicarbazones, although the overall molecular mechanism is still to be understood.
