Correlation between detection of a plasmid and high-level virulence of Vibrio nigripulchritudo, a pathogen of the shrimp Litopenaeus stylirostris

Vibrio nigripulchritudo, the etiological agent of Litopenaeus stylirostris summer syndrome, is responsible for mass mortalities of shrimp in New Caledonia. Epidemiological studies led to the suggestion that this disease is caused by an emergent group of pathogenic strains. Genomic subtractive hybridization was carried out between two isolates exhibiting low and high virulence. Our subtraction library was constituted of 521 specific fragments; 55 of these were detected in all virulent isolates from our collection (n = 32), and 13 were detected only in the isolates demonstrating the highest pathogenicity (n = 19), suggesting that they could be used as genetic markers for high virulence capacity. Interestingly, 10 of these markers are carried by a replicon of 11.2 kbp that contains sequences highly similar to those of a plasmid detected in Vibrio shilonii, a coral pathogen. The detection of this plasmid was correlated with the highest pathogenicity status of the isolates from our collection. The origin and consequence of this plasmid acquisition are discussed.

Amorphous-to-crystal transition in the layer-by-layer growth of bivalve shell prisms

Summary: Biomineralization is a complex process bio-controlled by living organisms, involving physical and chemical processes such as ionic concentration regulation and organic molecules production. This study investigates the early-mineralized calcareous units from two bivalve species using advanced optical and X-ray microscopy methods, revealing chemical and crystallographic insights. The observed ring-like structural features in calcite units are correlated with the absence of calcite and an increase in amorphous calcium carbonate and proteins content. Based on these findings, a temporal biomineralization cycle is proposed, providing biomimetic perspectives for designing crystalline materials.

SITVITBovis-a publicly available database and mapping tool to get an improved overview of animal and human cases caused by Mycobacterium bovis

Summary: Limited data on bovine tuberculosis and related infections are available. A publicly accessible database, SITVITBovis, has been constructed, providing genotyping and epidemiological data on Mycobacterium bovis and Mycobacterium caprae. With information on thousands of isolates from around the world, it allows for a wider understanding of the geographic distribution and phylogenetical sublineages. The database also includes molecular marker information and facilitates crosscheck among databases.

Systematic transcriptome analysis allows the identification of new type I and type II Toxin/Antitoxin systems located in the superintegron of Vibrio cholerae

Summary: In this study, additional type II and type I Toxin/Antitoxin (TA) systems were identified in the Vibrio cholerae N16961 genome. The functional type II TA system was encoded by the VCA0497-0498 operon, while the novel type I TA system was encoded by the VCA0495 gene and its associated antisense non-coding RNAs. These findings increase the number of TA systems in the genome and highlight their importance in this large cassette array.

Summary: Despite the complete sequencing of the genome of Candida albicans over two decades ago, there are still over 4300 uncharacterized genes. In this study, a collection of over 2500 overexpression mutants was constructed and evaluated to identify genes with fitness costs when overexpressed. This new resource enables routine large-scale screens under different growth conditions and facilitates the characterization of gene functions in C. albicans.

Summary: This study proposes a culture-based method with ertapenem selection to distinguish non-CRE, non-CP-CRE, and CP-CRE from samples with high bacterial loads. The method was successfully applied to clinical and environmental samples, allowing the identification of CP-CRE and detection of carbapenemase. This cost-effective strategy provides fast and reliable results and could be beneficial in resource-limited settings.
