Biochemical and molecular characterization of Cronobacter spp. (formerly Enterobacter sakazakii) isolated from foods

The aim of this study was to identify and characterize Cronobacter spp. isolated from a range of foods. A total of 71 Cronobacter strains were isolated from 602 foods in our laboratory. The highest contamination was observed in foods of plant origin, e.g. spices, teas, chocolate, nuts, pastries and vegetables. On the basis of genus and species identification performed using genus-specific PCR, 16S rRNA sequencing and AFLP genotyping, most of the strains belonged to Cronobacter sakazakii. Biochemical profiling by the tests included in API 20E, complemented with relevant additional tests, classified the strains into 13 biogroups. AFLP genotyping facilitated discrimination of six main groups at the 70% similarity level and strain grouping correlated clearly with species identification. Our results indicate that molecular typing by AFLP may be applied as a useful tool not only for direct comparison of Cronobacter isolates, providing traceability, but also for the reliable species classification. Moreover, tracing of these bacteria in a wider variety of foods should be important to enhance the knowledge of their transmission.

Impact of DNA extraction methods on 16S rRNA-based profiling of bacterial communities in cheese

Summary: This study evaluated the impact of different DNA extraction methods on the analysis of bacterial communities in cheese and found that various extraction procedures can lead to variable results, with the lysis method affecting the abundance profiles significantly. It also highlighted the risk of comparing bacterial community data in cheese when using different DNA extraction protocols and emphasized the need for a standardized approach for comparison across multiple sequencing runs.

Aspergillus niger Environmental Isolates and Their Specific Diversity Through Metabolite Profiling

Summary: This study presents a biological profile of 16 Aspergillus niger isolates from various environments, revealing high levels of toxic elements in contaminated soils and solid substrates. Molecular PCR and biochemical tests were used to confirm identification and characteristics of the isolates, with some strains capable of synthesizing mycotoxins. Environmental contamination may lead to genetic alterations in fungal species and influence their physiology, potentially resulting in the production of harmful metabolites.

Characterization and the host specificity of Pet-CM3-4, a new phage infecting Cronobacter and Enterobacter strains

Summary: In this study, four closely related bacteriophages belonging to Tevenvirinae subfamily were isolated, which infect Cronobacter and Enterobacter strains. Among them, bacteriophage Pet-CM3-4 exhibited broader host specificity towards Enterobacter hosts. Based on genome sequences, all these phages were assigned to the genus Karamvirus. Factors influencing the host specificity of Pet-CM3-4 phage and its host range mutant Pet-CM3-1 were studied, and a substitution in the long tail fiber adhesin was found to be responsible for the reduced host specificity of Pet-CM3-1. Furthermore, LPS was identified as the receptor for both phages, with C. malonaticus O:3 antigen being the receptor for Pet-CM3-1 and structures of the LPS core region being the receptor for Pet-CM3-4. These findings contribute to our understanding of the biology and evolution of Tevenvirinae phages.

Complete Genome Sequence of New Cronobacter-Specific Bacteriophage Dev_CS701

Summary: This report describes the characterization of Cronobacter phage Dev_CS701, a ubiquitous Gram-negative bacterium linked with serious infections, isolated from wastewater. Dev_CS701, related to phages belonging to the Straboviridae family and Pseudotevenvirus genus, contains 257 predicted protein-coding genes and a tRNA gene.

Bacterial and fungal communities associated with the surface of hemp seeds and poppy seeds

Summary: Microbial contamination of oil seeds can lead to deterioration and reduction of oil quality. Studying the levels and composition of microbial communities can help establish effective control measures. The bacterial community in hemp seeds is mainly composed of Enterobacteriaceae and Pseudomonadaceae, while the fungal community is mainly composed of Pleosporaceae, Davidiellaceae, and Tremellaceae. In poppy seeds, the bacterial community is mainly composed of Enterobacteriaceae (genus Klebsiella), Pseudomonadaceae, and Xanthomonadaceae or Streptomycetaceae. The fungal community is mainly composed of Pleosporaceae and Davidiellaceae (genus Cladosporium), with some samples showing increased levels of Trichocomaceae (genus Aspergillus). The presence of pseudomonads and aspergilli in certain seeds indicates the need for more effective cleaning and sanitation to prevent their impact on the produced oil.
