Summary: This study investigated the relationships among prodrug structures, pharmacokinetic characteristics, metabolic activations, and pharmacological responses of TFV ester prodrugs. The results showed that TMF and TAF exhibited stronger inhibition of HBV replication compared to TDF, and TFV ester prodrugs had a correcting effect on disordered hepatic metabolism. Improved pharmacokinetics of TFV ester prodrugs, especially intestinal stability, enhanced the inhibition of HBV replication and rebalanced hepatocellular metabolism. This research provides new insights and a basis for the design, modification, and evaluation of new TFV prodrugs in the future.

Summary: RNA-dependent RNA polymerase (RdRp) is an important drug target for inhibiting dengue virus (DENV) and has received attention in antiviral drug discovery. In this study, we screened 121 natural compounds from Litsea cubeba against DENV RdRp using computer-based drug discovery approaches. We identified four potential compounds (Ushinsunine, Cassameridine, (+)-Epiexcelsin, (-)-Phanostenine) with good binding scores and stable interactions with the target protein. Molecular dynamics simulation studies and post-simulation analysis were performed to assess the stability of the complexes and the drugs' interaction with the target protein. The findings suggest that phytochemicals derived from Litsea cubeba could be a therapeutic solution for controlling DENV infection.

Summary: This study designed and evaluated the combination of siRNA and liposomes to inhibit dengue virus replication. siRNA delivered through liposomes significantly reduced viral titers, without cytotoxicity, hemolysis, or stimulation of pro-inflammatory cytokine release. These results suggest that liposomal delivery of siRNA is a safe and effective method to inhibit dengue virus.

These drugs can inhibit virus replication or promote lethal mutations. Understanding the molecular interactions between these analogs and RdRP can facilitate the design of structure-based antiviral drugs. Clinical approval of some of these drugs highlights their significance in controlling viral infections.

Summary: This study introduces a DHE-based prodrug nanosystem (DHE@PEG-SS-DSPE) for ultrasound-driven tumor therapy. Under medical ultrasound, it produces cytotoxic ethidium and reactive oxygen species. DHE@PEG-SS-DSPE induces ferroptosis via glutathione depletion, lipid peroxide accumulation, and glutathione peroxidase 4 reduction. Furthermore, DHE@PEG-SS-DSPE nanoparticles not only exhibit effective ultrasound-active therapy of solid tumors but also downregulate the expression of metastasis-related proteins, inhibiting lung metastasis.

Summary: Nonlinear topological responses near exceptional points (EPs) greatly enhance the sensitivity of non-Hermitian systems to external perturbations. This paper proposes a new method to flexibly control the EP state of a whispering gallery mode microcavity integrated with photo-sensitive azobenzene coating. By introducing defects infiltrated with liquid samples, refractive index sensing can be achieved, and the proposed system exhibits higher sensitivity in nanoparticle detection compared to its counterpart operating near diabolic points.

By implementing a three-dimensional digital chaotic system, the security performance is effectively enhanced by scrambling the phase, symbol, and subcarrier frequency of the OFDM signal. The proposed noise-masking key distribution can transmit the key information with the chaotic encrypted signal, allowing uninterrupted authentication and decryption.

Summary: Introducing high-density defect sinks, such as grain boundaries and interfaces, to capture helium atoms and irradiation-induced defects is a promising strategy for enhancing radiation tolerance in nuclear materials. In this study, high-density grain boundaries and homogeneous interfaces were introduced into multi-nanolayered high entropy alloy films. The results showed that helium bubbles preferentially distributed along the grain boundaries in films with less interfaces, while introducing more interfaces led to the distribution of helium bubbles throughout the interfaces and a phase transformation. The underlying mechanisms of this behavior were explained and the roles of interfaces and grain boundaries in irradiation swelling and nano-mechanical properties were revealed.

Peer-to-peer (P2P) electricity transactions, as a new way of energy balance and interaction in the user side energy market, can promote energy sharing and improve economic benefits. Reinforcement learning (RL) is a suitable approach for P2P transaction decision analysis of small-scale users in the context of smart energy. The paper establishes a P2P transaction model and solves it using the SARSA RL algorithm, verifying the effectiveness, economy, and security of RL in solving energy storage decision problems.
