In Vitro and In Vivo Efficacies of Mefloquine-Based Treatment against Alveolar Echinococcosis

Alveolar echinococcosis (AE) is caused by the metacestode stage of the fox tapeworm Echinococcus multilocularis and causes severe disease in the human liver, and occasionally in other organs, that is fatal when treatment is unsuccessful. The present chemotherapy against AE is based on mebendazole and albendazole. Albendazole treatment has been found to be ineffective in some instances, is parasitostatic rather than parasiticidal, and usually involves the lifelong uptake of large doses of drugs. Thus, new treatment options are urgently needed. In this study we investigated the in vitro and in vivo efficacy of mefloquine against E. multilocularis metacestodes. Treatment using mefloquine (20 mu M) against in vitro cultures of metacestodes resulted in rapid and complete detachment of large parts of the germinal layer from the inner surface of the laminated layer within a few hours. The in vitro activity of mefloquine was dependent on the dosage. In vitro culture of metacestodes in the presence of 24 mu M mefloquine for a period of 10 days was parasiticidal, as determined by murine bioassays, while treatment with 12 mu M was not. Oral application of mefloquine (25 mg/kg of body weight administered twice a week for a period of 8 weeks) in E. multilocularis-infected mice was ineffective in achieving any reduction of parasite weight, whereas treatment with albendazole (200 mg/kg/day) was highly effective. However, when the same mefloquine dosage was applied intraperitoneally, the reduction in parasite weight was similar to the reduction seen with oral albendazole application. Combined application of both drugs did not increase the treatment efficacy. In conclusion, mefloquine represents an interesting drug candidate for the treatment of AE, and these results should be followed up in appropriate in vivo studies.

Summary: This study provides an overview of the prevalence and exposure to tissue cyst-forming apicomplexan parasites in Iranian domestic rock pigeons through molecular, serological, and histopathological examination. The results demonstrate that pigeons in Iran are infected, particularly with T. gondii, and suggest the need for control strategies to minimize the risk of infection in both pigeons and humans.

Synthesis and Antiparasitic Activity of New Trithiolato-Bridged Dinuclear Ruthenium(II)-arene-carbohydrate Conjugates

Summary: Eight novel carbohydrate-tethered trithiolato dinuclear ruthenium(II)-arene complexes were synthesized and evaluated for their activity against transgenic T. gondii tachyzoites. Seven conjugates strongly inhibited parasite proliferation while maintaining HFF viability. Two galactose-based conjugates (23 and 26) exhibited low IC50 values and reduced toxicity compared to the non-modified complexes.

Immunization with a Multivalent Listeria monocytogenes Vaccine Leads to a Strong Reduction in Vertical Transmission and Cerebral Parasite Burden in Pregnant and Non-Pregnant Mice Infected with Neospora caninum

Summary: In this study, a modified version of the attenuated Listeria monocytogenes vaccine vector Lm3Dx_NcSAG1 was tested for its efficacy in a pregnant neosporosis mouse model. Multivalent vaccines were generated by inserting gra7 and/or rop2 genes into Lm3Dx_NcSAG1. Immunization with Lm3Dx_NcSAG1 and Lm3Dx_NcSAG1_NcGRA7_NcROP2 resulted in the highest postnatal pup survival rate and protection against vertical transmission and CNS infection.

Summary: Gene knock-out using CRISPR-Cas9 is a useful tool to identify gene functions in T. gondii and other model organisms. Previous studies have shown that knocking out a gene can also affect the expression of other genes. Therefore, caution should be taken when studying the relationship between specific phenotypes and the knock-out of a particular gene.

Targeted and non-targeted proteomics to characterize the parasite proteins of Echinococcus multilocularis metacestodes

Summary: By using liquid chromatography tandem mass spectrometry, we identified parasite proteins in both vesicle fluid (VF) and vesicle tissue (VT), and found that the abundance of AgB subunits was the highest in VF from in vitro cultivation and in VF from a mouse model. We also demonstrated that AgB subunits from culture media (CM) can be taken up into VF within hours.

Summary: Artemisinin and its amino-artemisinin derivatives are potent antimalarials and exhibit different mechanisms of action against Plasmodium and T. gondii. The amino-artemisinins are effective in vitro against non-heme-degrading apicomplexan parasites including T. gondii. However, T. gondii strains readily adapt to increased concentrations of these compounds, resulting in the development of resistant strains.

Endogenous IL-33 Accelerates Metacestode Growth during Late-Stage Alveolar Echinococcosis

Summary: Infection with Echinococcus multilocularis can lead to alveolar echinococcosis, a severe parasitic disease. This study found that the release of IL-33, a signaling molecule, is associated with the development of the disease. IL-33 promotes the growth of the parasite and the progression of the disease, making individuals more susceptible to infection or disease. Understanding the role of IL-33 could provide new insights for the management and treatment of alveolar echinococcosis.

The Anthelmintic Activity of Praziquantel Analogs Correlates with Structure-Activity Relationships at TRPMPZQ Orthologs

Summary: The anthelmintic drug praziquantel is highly effective against parasitic flatworms, but its target has remained undefined. This study demonstrates that praziquantel analogs show different activities against different parasites, which is reflected by unique structure-activity relationships at the TRPMPZQ channels found in these organisms. These findings provide further support for TRPMPZQ as the therapeutically relevant target of praziquantel.

Establishment and application of unbiased in vitro drug screening assays for the identification of compounds against Echinococcus granulosus sensu stricto

The study provides reliable in vitro culture techniques for generating E. granulosus s.s. metacestode vesicles and GL cell cultures and validates standardized in vitro drug screening methods for E. granulosus s.s.
