Inhaled corticosteroids reduce senescence in endothelial progenitor cells from patients with COPD

Summary: Cellular senescence is accelerated in smokers and COPD patients. In COPD patients on inhaled corticosteroids, cellular senescence and DNA damage response are reduced. In vitro studies suggest that corticosteroids protect endothelial cells by reducing oxidative stress-induced senescence and apoptosis.

Autoimmune disease and COVID-19: a multicentre observational study in the United Kingdom

Summary: Patients with autoimmune disease (AD) admitted with COVID-19 showed differences in laboratory findings and clinical outcomes compared to patients without AD. Those with severe rheumatologic AD had higher mortality rates and elevated levels of anemia, renal impairment, LDH, CRP, which are associated with increased mortality in COVID-19 patients.

Summary: Antiphospholipid syndrome (APS) is an autoimmune disorder characterized by vascular thrombosis and pregnancy morbidity. The current treatment options have limitations, and there is a need for more targeted therapies. Researchers developed a modified protein variant using site-specific chemical addition of polyethylene glycol (PEG), which effectively inhibited thrombus formation caused by pathogenic antibodies. These findings provide further evidence supporting the potential efficacy of PEG-modified protein as a treatment option for APS.

Summary: This study investigated the outcomes of children born to mothers with SLE exposed to HCQ or AZA during pregnancy. The results showed that there were no significant negative outcomes in children exposed to HCQ during pregnancy. However, the use of AZA was associated with an increased reporting of childhood infection, which requires further study.

Antibodies to FXa and thrombin in patients with SLE differentially regulate C3 and C5 cleavage

Summary: The significance of antibodies directed against activated factor X (FXa) and thrombin (Thr) in patients with SLE and/or antiphospholipid syndrome (APS) is unknown. This study found that aThr IgG increased Thr-mediated activation of C3 and C5, while aFXa IgG did not increase C3 or C5 activation. Structural analysis suggested that aFXa IgG may have a higher likelihood of steric hindrance on FXa compared to the AT-Thr-aThr IgG complex. Although there was no significant association between positivity for aFXa or aTHr IgG and C3 levels or disease activity, there was a trend for patients positive for aFXa IgG alone or both aFXa and aThr IgG to have lower levels of C3.

Hydroxychloroquine as an Immunomodulatory and Antithrombotic Treatment in Antiphospholipid Syndrome

Summary: Antiphospholipid syndrome (APS) is a highly prothrombotic disorder caused by thrombo-inflammatory antiphospholipid antibodies (aPL) that lead to thrombosis through various mechanisms. Obstetric complications in APS are a result of placental thrombosis, inflammation, and complement activation. While anticoagulation is the main treatment, it may not effectively prevent thrombosis recurrence in some patients, especially those with triple positive aPL.

Evolution of Antiphospholipid Syndrome

Summary: Antiphospholipid syndrome (APS) is an autoimmune disease characterized by the presence of antiphospholipid antibodies, leading to thrombosis and/or pregnancy complications. APS has a long history dating back to the mid-20th century, when it was first identified during syphilis screening programs. While anticoagulation with vitamin K antagonists remains the main treatment for thrombotic APS, newer options like direct acting oral anticoagulants have shown inferiority, particularly in patients with triple-positive APS and arterial thrombosis. Immunomodulatory agents such as hydroxychloroquine are increasingly used to treat refractory cases of APS.
