The occurrence of psoriatic arthritis in Denmark

Objective: To apply and compare different classification criteria on a representative nationwide sample of psoriatic arthritis (PsA) twins and to estimate the prevalence and incidence of PsA. Methods: The study comprised three Danish nationwide twin cohorts. In 1994 37 388 Danish twin individuals and in 2002 46 418 twin individuals received a questionnaire, including questions on rheumatic diseases. Twins reporting PsA and their co-twins were classified according to the Moll and Wright and CASPAR (CIASsification criteria for Psoriatic ARthritis) criteria based on interview, clinical examination and scrutiny of medical records. Results: 228 twin individuals reported PsA and 164 (72%) participated in clinical validation. By using the Moll and Wright and CASPAR criteria, 54 and 50 cases were diagnosed with PsA respectively. The positive predictive value of self-reported PsA was 31%. According to the Moll and Wright and CASPAR criteria the prevalence was 0.15% (95% Cl: 0.13%, 0.22%) and 0.14% (95% Cl: 0.11%, 0.19%) respectively. The annual incidence rate based on new self-reported cases in 2002 was 6/100 000 person-years (95% Cl: 3/100 000 person-years, 11/100 000 person-years). Conclusions: The positive predictive value of self-reported PsA was 31%. The prevalence and incidence figures of PsA were equivalent to the previously reported occurrence in population- and hospital-based studies.

Summary: This study conducts a genome-wide association analysis of venous thromboembolism (VTE) using a large cohort of cases and controls. They identify 93 risk loci, with 62 loci being newly discovered. The study also demonstrates the effectiveness of a VTE polygenic risk score in identifying individuals at high or low risk for VTE. Additionally, they investigate the shared clinical risk factors between venous and arterial thrombosis.

Cluster headache polygenetic risk and known functional variants of CYP3A4 are not associated with treatment response

Summary: This study investigated the association between polygenetic risk, functional variants of CYP3A4, and treatment response in cluster headache patients. The results showed that the response to oxygen, triptans, and verapamil was associated with the chronicity of cluster headache, but not with a high genetic risk. The response to verapamil was also not influenced by functional variants of CYP3A4. Furthermore, no support was found for the genetic variants previously reported to be associated with treatment response. Overall, this study suggests that the clinically relevant variation in treatment response in cluster headache is not influenced by genetic factors.

Rare Heterozygous Loss-of-Function Variants in the Human GLP-1 Receptor Are Not Associated With Cardiometabolic Phenotypes

Summary: This study identified 36 GLP1R variants in Danish individuals, of which 10 showed a loss of GLP-1-induced signaling. However, no association was found between these variants and type 2 diabetes, although carriers exhibited slightly higher fasting plasma glucose levels. Additionally, the analysis of UK Biobank participants did not reveal substantial associations between GLP1R variants and cardiometabolic phenotypes.

An adult-based genetic risk score for liver fat associates with liver and plasma lipid traits in children and adolescents

Summary: This study investigated the effects of eight previously reported genome-wide significant steatogenic variants on liver and cardiometabolic traits in children and adolescents. The results showed that these variants were associated with higher liver fat and distinct patterns of plasma lipids. The genetic risk score (GRS) based on these variants was also associated with higher prevalence of hepatic steatosis and had potential clinical utility for risk stratification.

Summary: This study investigated the role of gut microbiota in the pathogenesis of anorexia nervosa (AN) by analyzing metagenomics and metabolomics data from AN patients and healthy females. The results showed that AN patients exhibited alterations in gut microbiota composition and function, as well as increased levels of metabolites associated with reduced food intake. Microbiota transplantation experiments further confirmed the contribution of gut microbiota to AN behavior.

Circulating microRNAs for Early Diagnosis of Ovarian Cancer: A Systematic Review and Meta-Analysis

Summary: In this study, a systematic review and meta-analysis were conducted to evaluate the global research potential of different circulating miRNAs as an early diagnostic biomarker for ovarian cancer (OC). The primary search resulted in 1887 articles, and 44 relevant studies were identified. Nine miRNAs were found to be dysregulated in OC patients compared to controls, with miR-21, -125, -141, -145, -205, -328, -200a, -200b, and -200c being upregulated. Future studies of circulating miRNAs in relation to OC should consider factors such as sample size, consensus guidelines, and coverage of previously reported miRNAs.

Summary: This study aimed to develop protein risk scores for ASCVD risk prediction and compare them with clinical risk factors and polygenic risk scores. The results showed that the protein risk scores were significantly associated with ASCVD events and provided a modest improvement when added to the clinical risk factor model.

Summary: In this study, we investigated the prognostic accuracy of preoperative plasma suPAR in patients with localized ccRCC. The results showed that elevated suPAR levels (≥6 ng/mL) were associated with a higher risk of disease recurrence and lower overall survival. suPAR could be a promising surveillance tool for monitoring ccRCC patients and detecting future recurrences.
