Summary: The synthetic utility of heterotelechelic polydimethylsiloxane (PDMS) derivatives was enhanced by using a specifically designed silyl hydride (Si-H)-based initiator for anionic ring-opening polymerization (AROP) of hexamethylcyclotrisiloxane (D-3) monomers. The resulting PDMS polymers had high chain-end fidelity and heterotelechelic functionality. In situ termination of the propagating chain end with different chlorosilanes and functionalized alkenes further increased the diversity of the PDMS derivatives. The living nature of the polymerization allowed for the synthesis of narrow-dispersity polymers with a wide range of molar masses. Additionally, the feasibility of synthesizing supersoft bottlebrush networks using the PDMS macromonomers was demonstrated.

Summary: Every type of cancer tissue is more susceptible to viral infection, which has been utilized as a new anti-cancer therapy using oncolytic viruses (OVs). OVs selectively infect and destroy cancer cells while causing no harm to the host. Research focuses on the interaction between OVs and the host immune system, particularly the role of dendritic cells (DCs) and natural killer (NK) cells in the therapeutic response.

Summary: We have developed a novel biotechnological platform called MAGIC, which overcomes the limitations of current conjugate vaccine design methods and improves the yield of glycoconjugates. We have also improved MAGIC to allow rapid installation of glycoengineering components, eliminate the use of antibiotics, and reduce reliance on protein induction agents. Additionally, we demonstrate the versatility of the MAGIC platform in performing glycoengineering in genetically less tractable bacterial species. The MAGIC system promises to be a rapid, robust, and versatile method for developing vaccines against serious bacterial pathogens.

The results show that EAV is able to replicate in monocytes and mature MoDC, and has an effect on cell morphology, phenotype, and function. EAV evades host anti-viral immunity through inhibition of antigen presentation early after infection and by killing infected DC during replication.

Summary: Fluoro-poly(acrylic acid) heterotelechelic homopolymers have been synthesized to mimic the ultrafine domains of high chi-low N diblock copolymers. The thermodynamically stable nanomorphologies of lamellar or hexagonally packed cylinders were obtained by heating/cooling ramps. These telechelic homopolymers mimic high chi-ultralow N diblock copolymers and enable reproducible targeting of nanomorphologies with incredibly small, tunable domain size.

Summary: The study demonstrated the in vitro priming of naïve human T cells against heat-killed B. pseudomallei by co-culturing with syngeneic, pulsed dendritic cells. Myeloid DCs showed a major response to B. pseudomallei, contributing significantly to the expression of migration marker CCR7 and secretion of proinflammatory cytokines TNF alpha and IL1 beta. The study suggests that this approach could be used to screen for human cellular response to candidate vaccines and enhance cell-mediated immunity against intracellular pathogens.

Summary: Recent estimates show that one in two sexually active individuals will acquire a sexually transmitted infection by age 25, highlighting the importance of vaccination against STIs. Vaginal immunization is a promising strategy to combat transmission via the vaginal mucosa by overcoming immune tolerance and establishing tissue resident memory immune cells. Research is focusing on exploring the potential of vaginal vaccine studies and the role of mucosal adjuvants and delivery systems in enhancing immunity.
